Clone vs MS, Clone Selection
    #11304670 - 10/23/09 09:19 AM (7 years, 11 months ago)

Will cloning the mycellium from a fruit and using that mycellium for new fruits result in a more prolific fruiting strain/flush/pinset etc... 99% of the time vs multispore?

When selecting a clone should one look for the biggest fruit or the biggest cluster (E.g. clone big fruits = many big fruits, clone cluster = many clusters?)

Is it better to harvest clusters or pins for cloning when they're young or let them mature?

Is there any point to take multiple tissue samples from a single fruit for different LCs?

Is the best route to clone a number of promising specimens then grow them all out and use the clone that worked best?

Once you have this best working clone should you just use that myc or attempt to clone a specimen again to further refine traits? If one takes a spore print of this fruit grown from clone genetics will the print carry the same traits or does it need to be further isolated over and over?

I remember reading that commercial edible mushroom farms would find their fruiting strain by cloning pins of clusters (maybe doing this repeatedly) so that theyd have full canopy pin sets

Usually.
Your choice.
Your choice.
Not really, but no harm either.
Yes.
Clone = final product. Don't reclone.

Cloning clones isnt such a good idea from what ive read a copy of a copy of a copy, takes down the potency, like b+

is it the senescence factor?

I dont buy the potency loss part.:stoned:

spore prints carry millions of different traits, by cloning it before printing it vs. not cloning first wouldnt really make that much of a difference, if you wanted to clone a specific mushroom and get the same traits over and over again, make an Lc master, then transfer a few cc's, to make a working Lc, store your master in the fridge if you want to halt growth for a while. or work with agar, it too can be stored in the fridge for some time, im not exactly sure how long thou, you would need a more experianced grower to tell you that
I seriously hope this helps even if it only helps a tiny tiny bit
Peace

nothing personal AIM, just trying to unconfuse myself.

I know how to clone/isolate and all that jazz, Im mearly wondering why its not advised to clone clones.

is it due to senescence because its the same myc network continued or is it something Im missing?

this is the only thing close to an answer I can find.

age would be the issue, but its easily solved.

you can clone clones just not to many times.:stoned:

Thats what I was thinking and makes  a lot more sense. If you truly were to suffer genetic recession in performance due to cloning you could just use the mycellium from the previous generation that wasn't defective.

I remember reading about commercial edible farmers finding their strain by cloning clusters of pins, and cloning the clusters of pins of that clone and so on until all they had was full canopy. I imagine it would be quite feasible to get a stable prolific fruiting strain within a couple generations of cloning before suffering a setback in performance from the cloning.

Im no mycologist but the idea that cloning a clone can never be beneficial in refining the desired traits doesnt make sense to me

If you store the cloned tissue in a test tube (like the ones in RR's video, with agar and a piece of wood) u don't need to reclone, whenever u feel like growing that selection again, just use a small bit to make a LC. The test tube can be stored in the fridge for years and you always can transfer to a new test tube later.

Senescence is exactly why you don't want a tissue culture that is "too" old.

Ideally you want to isolate directly from spores, so that the master culture is maintained as youthful as possible, but it's a consuming process to get desirable traits. Cloning pins let's you get a culture that has certain traits and is still relatively young. Pins are one of the most rapidly dividing types of mycelium so they make good cloning material.

Re-cloning a culture down the line doesn't make much sense... it seems like just getting the same sample of a culture when it's that much older.

I haven't figured out how to get around the quote thingy, but do you have the link to the video still?
