Spores under a microscope
    #11260819 - 10/16/09 05:49 PM (8 years, 8 days ago)

Got a microscope today. And my spore prints are ready too. So, logically, did the thing they were bought for (the place I got them sells them explicitly as spores for microscopy )

P. cubensis "Cambodian" at x1000 magnification. Enjoy. And post yours! Is there a thread or database of spore images?

Nice, you can see that the spores are collapsed, to view them better you should let them hydrate in a drop of water on the slide before placing the cover slip over them, I find leaving them for half an hour or so in water sufficiently hydrates them!

You can germinate them on the slide too.  This is 400X.

Nice shot Roger, do you add some form of sugar to the water for germination, I have seen it happen with plain water but maybe honey water would work better!

Thanks for the hydration tip, I assumed them being collapsed was a feature of the spores.

I will definitely try to soak them in honeywater to see if that promotes germination.

Do you use a thin top cover glass above the specimen, or can that hinder possible mycelium growth?

Karo is better because it's clear.  It doesn't cloud the slide the way honey does.  With very fresh spores, you'll start to see germination within twenty minutes to an hour.

You have to use a cover slip.  Otherwise, your spores would not all be in the same plane of focus.
