Fractional sterilization

Will fractional sterilization work to sterilize grain? has anyone had success with this method?

I tried several times to sterilize rye berries and then birdseed with this method. Every attempt ended in contamination.

well I havn't tried it but I would:

boil 30 min
incubate for 24 hours at 30C
boil 30 min
incubate 24 hours at 30C
boil 60 min

And the signature is just a pearl of wisdom I had whe I was high once lol

I was joking, sorry if it didn't sound like it.  I was just playing.  :tongue:  But I still can't understand it.  I'm serious, I've read it when I was sober, stoned, drunk, a combo of the last two, and I can't get it, damn damn damn.  Oh well.  ok, back on to fractal sterilization. :wink:

It's not supposed to make sence. You can't drive a drink, a drink cannot be driven. I just find it funny when I read it. If you ever actually understand it, you must have like 20g of shrooms in you.

For the rye berries I first simmered the rye at a low boil for 40 minutes. I then dumped the rye into a strainer and let it drain. After it was well drained I loaded it into a quart jar 2/3 full, put a filter disk on it, covered the disk with a lid that had two 3/8" holes drilled into it and then secured it all with the ring. I covered the filtered holes with duct tape, boiled the jar for 1 hour and then let it sit in the pot for 24 hours at about 85F. I repeated this three times. On day four I removed the jar from the pot, removed the tape from the filtered holes and then inoculated through one of them. The tape was then replaced over the inoculation hole, leaving one filtered hole for air exchange. I'm wondering now if I might have had better luck using smaller jars so that the heat could penetrate more easily.  :confused:

When using grains as a substrate a pressure cooker is a must.

Dog your problem might have been your incubation temp. You had it at 85 when it should be between 86F - 99F. I'm going to try this using a pint jar and a tall 1/2 pint jar. I'll post my results when I do this which willl be in the next 3-4 weeks.

The 85F was to encourage germination any endospores residing in the grain so that they could be killed in the next round of boiling. 85F is also ideal for incubating cubensis. Check out this chart from the FAQ:



If you try to incubate at much more than 88F you are going to be waiting for a fuck of a long time.

when I said your incubation temp should have been between 86 - 99 I meant while incubating the endospores during fractional sterilization not after inoculation, I thought that was pretty obvious. your incubation temp (for the endospores!) of 85F was probably too low and didn't cause all of the endospores to germinate, therefore letting them survive the boilings

Read lots of teks on sterilization And do exactly as they say and maybe then some if not whats the point in trying alot will get contamed its the number step and most important in my words

I did the same procedure you described, and the rye jars turned out perfectly, contaminant free and colonized completely in less than 2 weeks.
