Pasteurizing substrate in filter patch bags
    #11244832 - 10/14/09 09:33 AM (14 years, 5 months ago)

I couldn't find a definitive answer anywhere, so if i overlooked it, please just post a link to the thread
Here's my question:

I have large autoclavable filter patch bags. Wondering if I can pasteurize horse poo/verm/straw, whatever in the bags (using a stovetop roast cooker)
I would then innoc. the pasteurized bags with colonized WBS in a glove box, seal with tie wire, and let fruit in the bags. Would this work?

Yes, I do it all the time.  Stick a meat thermometer into the center of the substrate and heat the water the bag is sitting in until the substrate has been at 140F to 160F (60C to 70C) for one hour.

I am doing this right now with 2 bags

Thanks a lot. How necessary is it to fill up the bags with sterile air (as per your DVD)? I don't have a flow hood, just a pretty good glove box.

Sterile air is kind of important when your doing an in vetro grow. The reason why is because any nasties that land in there will absolutely LOVE the growing environment that you have provided for your mycelium.

Once the mycelium has taken hold, you don't have to worry as much.

You really don't have a choice other than working with what you have got. Just be as clean as you possibly can. Use your glove box. Take all the precautions that you can.

Remember that other growers have been successful with less than you have. (This will keep you from having an 'I can't be successful' attitude).

Maybe put a syringe filter on a big ol' syringe and inject air into it?

buy a tyvek painting suit or get it for free from the post office, and use tyvek for a breathable filter. Dont mess around with injecting air into it manually

They're already filter patch bags. What i'm curious about is if i should provide more oxygen as the bag colonizes.

They dont need lots of oxygen to colonize they just need Gas exchange. They need fresh air exchange when there ready to start pinning and to fruit

if you hang the bags, they will draw air through the filter patch until pressure is equal in and out of the bag. once pressure is equal, you have enough air in the bag

are you confusing what steps require pasteurizing and which require sterilizing?

I was talking about what RR said in his DVD
He said to fruit in vitro you need a lot of (sterile) air in the bag.

you want to pasteurize your bulk substrate that you will be spawning the colonized mycelium to.

you want to sterilize whatever it is you choose to inoculate with the spores.

it just seems like you are confusing the two throughout the thread.
