cleaning up spore prints
    #11227298 - 10/11/09 05:26 PM (8 years, 9 months ago)

alright so i been doing this for a while but this is a process i never really had to look into so never got it down ... Ive recently come across a copelandia infestation in one of the areas i hunt and i want to get some prints but considering the environment they came from it seems rather unlikely ill succeed in pulling off anything sterile .... i have heard tell of a remedy for this though , just cant really pin down the specifics ...

Spore prints are never sterile. The mushroom itself is exposed to non-sterile environments when it's being fruited.

If you want to clean up a print from the wild, agar would be the best route to pursue. If you are having a bacterial problem that anti-biotic agar or MEA can't handle, try using getamycin in your medium. It was recommended to me by RogerRabbit and is also referenced the Paul Stamet's Book, GGMM (Growing Gourmet and Medicinal Mushrooms).

I'm not really familiar with copelandia, or tropicalis... but have you considered trying to take a tissue culture from your outdoor samples?
you can clean the outer tissue, and cut it open to remove a chunk, and either move directly to agar, or even try a liquid culture. and by taking a tissue sample, you can then use peroxidated agar. giving you a jump on getting ahead of any contaminants, while not destroying the mycelium.
if your only looking to get prints from them, then of course, chances are you will have several transfers to do before you can get a cleaned up sample. doing a tissue culture on the other hand, you get the chance to wash/clean your sample, and the tissue from the inside of the mushroom will be clean anyways, provided you're sterile while collecting your tissue.
just some ideas for you to consider. GL with you find

I second Jitsu.

do agar transfers until you have a clean isolate or use antibiotic agar w/gentamycin to really hedge your bets.
