GRRRR 100% contaminated AGAR again..

ok i have wasted so many dishes...

i mixed agar and water and nutes (molasses, it works)

i PCd the mix for like 20 min

i poured into dishes while still hot.

3 days later all the dishes have little spots all over the agar, little circles of contam growth..
GHRRRRRRRRRRRRRRRRRRRR

what did i do wrong????


im having so much trouble with agar..

How are you pouring the dishes? Glovebox? How sterile? Gloves? Mask?

well i was pouring in the open, that i guess is the issue.. i only opened the dish enough to pour but i guess that i need to do it in a glove box...

i wish there was an easier way..
like a BIG syringe full of sterile agar and trays with self healing ports for filling... that would make it alot easier..

i've been having somw bad luck with my agar a little lately also even with glovebox, gloves enough alcohol to kill a horse and Oust oh well keep on truckin...

Maybe the molasses is too rich in nutrients. I have heard a lot of people who use agar shun the use of high sugar products in agar such as molasses and honey because it is too contam prone. I just did like 15 grams of potato flakes with 10 grams of agar and 500 ml water and it worked pretty well.

Agar is super-sensitive. There are places where you can handle it in open air, but I don't think most of our apartments qualify.

People have used 1/4 pint jars for agar, you can use a regular lid with it.

No need for GE on the 1/4 pints? j/w cause I'm using some 1/4 pints right now that have some GE holes but would like to start some more and if I dont have to make GE holes on the other ones it will make it alot easier.

If you're messin with agar then you'll NEED at least a GB anyways. Rig one up ... it doesn't have to be anything special.

i have one.. just made it 2 days ago.. but did not have it when dishes were poured

i just nuked the dishes till the agar boiled again.. that should kill most of the contam on the plates..

ill let ya know how it goes.
