weak syringe solution?

Of 12 PF tek jars, about half of the innoculation points are showing NO growth whatsoever a week later. Each 1/2 pint jar had 4 innoculation points. Some are showing strong mycelium growth and a few are showing very weak growth (which may be cobweb) and the rest look exactly how they looked after sterylization. I know it doesn't necessarily mean much, but the syringes were very clear and had hardly any black floaties. Did I get ripped off?

Did you shake the hell out of the syringe before each innoculation? You should have.

Spores are microscopic so a clear syringe means nothing.

injected more than the reccomended 1/4 cc. of solution per innoculation point. not too concerned, its just that the whole process will take a little longer now. just wondering why some innoculation points have developed while others haven't? made sure the syringe solution was mixed well before each innoculation. what factors may have caused this?

I made a syringe this morning, and it doesnt have any visible specks. It was from a very small PR print on paper. Very tough to scrape that little thing, but Im sure there are enough spores for my jars :smile: I HOPE! :cool:

Your substrate could be a factor. What recipe did you use?

Patience - I've found that some colonize considerably slower than others from the same group for no apparent reason...I have a pink buffalo that just sat there with moist spots for almost a month (!), and i just left it there kinda out of curiosity, and now it's doing fine. I think I may not have tamped it down enough when I prepared the jar, which might have caused the mycelium to struggle trying to bridge gaps in the substrate as it establishes itself.

If you have an innoculated jar without any growth you might wanna give it a smell. Ewe, sour apple!

Temperature is a pretty big factor during colonization. Try and keep it around 80 degrees.

In my experience, I have found that when I try any extra water with the PF tek, colonization slows considerably. The mycelium which is usually a bright white, appears to grow a dingy grey, and is much less prominent. Eventually when the water is used up a little bit, the mycelium establishes a better grasp on the substrate and will eventually look normal. How much water did you use? Did you mix up your substrate for individual jars or in a bulk mix. This impacts the water content of some parts of the substrate. Maybe these bad jars are water lacking jars or have extra water, perhaps too much.

I have found that the solution for problems like this... at least in my configuration, is the time of incolation, Unshaken Suringes, Make sure you see incolant hit the side of the jar, i would reincolate after another week or two if no myc

the 12 jars are in a "rubbermaid-in-rubbermaid" incubation chamber (the lower container w/ heated water). the ambient temp. of the chamber of the chamber is generally around 82 degrees. the syringes were shaken thoroughly before each individual inoculation and the needle was pressed against the glass. about 2/5 cc was used per inoc. point--- what is strange is that, of the total 48 inoculation points, most have growth, but about 14 points are showing NO growth....the puerto rican jars are inoculating much faster than the B+. really don't know what the problem is, but probably won't order spores from this vendor again. thanks for all the help.

i have considered that the growth-less jars were perhaps too dry but looked at them under the light today and saw alot of condensation on the inside of the jars.
these jars were re-innoculated the with PR solution (which has been more succesful than B+), so hopefuly there will be results in a week or so.
also, some of the jars do have more water than others, perhaps TOO much and, as you refered to, the mycelium growth is "dingy gray" and much less impressive. i've learned that you can read as many teks as you want, but you must gain first-hand expeience in order to improve at this hobby. thanks
