First Bulk Grow in Trouble...
    #11194184 - 10/06/09 02:27 PM (14 years, 6 months ago)

Hope everyone is doin absolutely swell today. I could really use some advice from some of the experts here since I'm doing a lot of teks for the first time so I'm not sure how well it was executed.

I don't know why but I feel rushed when I prepare my shit. I can't take my time even though I'm operating under ideal conditions. Maybe its a fear that EVIL spores are floating around to sabotage my grow :mad2:

Meet my shitty LC:

While I did succeed in growing some mycellium, it really spread out and hard to capture in the syringe. Do you think there are traces of myc everywhere and that I don't need to actually suck it up the visible white into the syringe?

I'd like to prepare some that was thicker. I used the Karo Tek using about 5% syrup per volume.

Meet my WBS:

To prepare my WBS I synthesized the 6T Tek and Doc's Tek meaning that I soaked 13 hrs, picked out the 'floaters' but didn't simmer before loading the jars in the PC.

Things I noticed:
1. The WBS mildly sticks to the sides of the jar. (1st picture)
2. It seems to have discolored and is now darker than what I see here on the forum.
3. Its been incubated for 4 days and still no growth even though it was inoculated with live culture.

What do you guys think? Any LC suggestions? Should I just keep a watch on the jars? General advice?

Thanks a ton.

WBS looks ok, maybe a bit wet.
LC looks lame. You didn't say how long you let it grow, but it wasn't long enough by far.

Did you make your LC in a glovebox or in front of a flowhood? I find it impossible to inoculate any liquid medium and get ONLY mycel in open air. To ensure the mycel is evenly distributed through the entire mixture I keep a glass bead (an agitator) in my liquid cultures. Just a small glass bead in the baby bottles. The agitator breaks up the mycel when you stir/swirl the LC, for when you draw in LC solution into the syringe.

Here is another WBS prep method. Foo Man's WBS Method

The LC has been growing for about 2 weeks now.

I second the wet appearance of the WBS but I don't see how it got wet when I let it dry for 2 hours. Maybe it got in through the loosened jar lids.

You should boil it for better draining. You don't have to simmer, just bring it to a boil and turn off.
You started your LC from spores I guess? It might take off, it might not. 5% is a bit rich, try 3-4% next time too.

Thanks Doc & Others...

Can I get confirmation that I should be seeing some growth now if I inoculated it with live LC last Saturday?

Thanks.

Starting with live LC, you should see growth pretty quickly. It may take it a day or so to adjust to a new rich food source, but a healthy LC should just take off from there.
If yours hasn't... give it some time but have a backup plan.

My backup is soaking at the moment. This next time will be different. Doc, I'll be sure to let you know when I finally get this right. Thanks again.

Do it the other way around, make Grain LC from colonized WBS. Works about a thousand times better.

I agree with Doc. If you get a jar of colonized grain that is healthy and COMPLETELY colonized you will be so happy with the results of a grain lc. even if you don't do cakes, you should try one or two (just adjust the basic recipe) to see how very aggressive it is. You can even do a pint or whatever you have available for the grain but you will want a quart. The principles are the same. Just remember this. I have found that I always want to prepare extra syringes of sterile H2o for the grain lc process and have them ready and standing by. I can never get back in water what I put in. I also can tell you that you might want to try the high temp RTV style lid thingy for your jar of grain LC and put your RTV injection point nearer the edge of the jar than in the middle for this kind of tomfoolery. It makes it sooo much easier.
There is a great tek for this. Listen to the good Doc.

Thought I would mention...

I tried simmering the WBS last night. Apparently I over did it because all the grains were popped and I was left with a sludge. :crazy2:

I swear, I am having the worst luck with this. WBS must be picky as hell. I've wasted so much of my time and energy already.

Bring it to a boil, turn it down. Cook about 5 minutes- use a timer. Then eat a kernel once a minute. Use a timer.
When the grain is no longer crunchy- but before it gets squishy- drain the hot water off of it quickly and let it start cooling.
Remember how long it took, set your timer to that the next time. But still eat a kernel to make sure it is al dente.

Oh, and it helps if you use a timer.  :getstoned:

The human mouth/nose complex is one of the most sophisticated analysis instruments conceivable.
You can analyze grain for moisture content instantly with perfect accuracy, simply by eating a kernel.

I don't taste everything, but I'll try it if there's a reason. Haven't tasted gypsum yet, for example.

good answer... when i was still in school, working in this lab, people thought it was fucking disgusting when i started taking sips off the agar mediums i was preparing...  they already thought the smell was bad enough, then me rubbing it in their faces by drinking the shit... i thought it was hilarious.  I've tasted plenty of gypsum though, i've done my fair share of drywall hanging and finishing...

i don't usually chew on the kernels to determine moisture content, usually can figure out just by squeezing them between thumb and index finger, but that does't mean i haven't eaten them before...
