Getting Frustrated: Contams...contams...contams

One three occassions now I have purchased pre-made rye jars and wbs jars from reliable sources only to have them contaminate upon inoculation. I've tried multispore syringes...then made up LCs (which clearly work based upon their "spermy" content), but regardless of the source, I get contams...typically "wet spot" bacillus or aspergillis.  I've tried different strains...KS, Cambo and B+.  My asceptic technique is good enough to work with agar...I flame the needle..I wrap in alcohol soaked cotton...I flame after each inoculation.  I shower first....I wear clean clothes.....I Lysol the room in which I work, I alcohol all the surfaces.  Still..simple innoculations into purchased jars always seem to result in contams.  My PDA petri dishes look good...but not my jars.  I've built a TiT..I have a glove box and a nice looking FC.

I started working in the Spring...and I still havent gotten a single mushie. I read every tek and log on Shroomery which always drives me insane with jealousy since people are always talking about their half pound yields (wet of course)...and I cant seem to get a single jar to fully colonize to that white white white color...let alone spawn it to hpoo and coir. 

I obviously need to either quit trying to grow and buy my mushies from some kid in the hood, or need some real help in getting a grow together. Maybe its the commercial jars I am buying? Can anyone recommend the best supplier for these?

Any suggestions would be appreciated. I guess I could go back to BRF cakes. With those I would at least get 50% to be usable.  That really feels like admitting defeat...but I do want some mushies again.

make your own jars cheaper and better. that is the first place to start. if it works on agar then the jars are the problem. how much solition are you putiing in the premade jars? what size are they? get from a vendor of this site? i assume you didnt cuz if you did you wouldnt have this problem

1) Can't tell anything by looking at LC.
2) Premade jars? Make your own.
3) Have you tried an agar wedge into one of the premade jars?

1 again: If you are using untested LC to knock up your jars, I guarantee that's the problem. How are you making it?

So..you think its the commercial pre-made jars?  There must be a reliable vendor out there who actually pcs their jars as long as they should.

My LCs are karo syrup and water.  Doc...I've plated a few drops out on PDA agar before using them to inoculate.  I only use the LC if it grows myc on the plate.

or the LC since you dont know if it is good till you put on agar or inoculate but since you dont even know if the jars are good it dosent matter

There you go. If you make the agar and you make the LC and the LC you make works with the agar you make... the grain jars are the problem. (Always test every LC, don't just assume that they are good.)

How much are you shooting in?

I inject about 4 ccs of the LC into each jar.  I try and make sure that I can see mycelium floating around in the syringe before injecting.

I am hearing everyone saying that its the commercial jars and I will do better with making up my own jars or going back to BRF cakes.  *sigh* I know you guys are right. I am spending so much on the commercial jars...that I could have bought two PCs by now.

At least you can empty out the jars and reuse them. Are the lids nice?

man man man got contam........

i had this happen too..... if the plate is good and you are getting success with that then ya its the jars. but still could be the lc. next lc batch do an extra control jar and put nothing in it. that way you can see what it is supposed to look like with no contam. knock up the remaining lc jars with agar wedges in the g-box. be quick and so forth. in 4-5 days compare all the jars. the ones that are milky ad not clear are contamed. the ones that are still clear and you see myco groth / wispy things are good. and you can move on to the jars.

well do the same thing leave one un knocked and keep it withe others as a control.

good luck and by the way this is what i do too. just to be safe and give me an idea were the problem is.

if you are makeing lc i assume you have a pc so getting to a feed store is the next step.

Yeah...the lids are very nice!! Tyvek and an injection port...and I DO have a bunch of them now.
Ratdog...I like your experimental approach to the problem...a good control is always informative. I've used agar wedges to knockup a commercial half gallon rye jar...which grew quickly and then developed green patches...and out it went.

that sucks about the green thing guy. but i too have that happen so i do several jars and when i get 100% in the good jars i do g2g in larger jars. just seems faster that way. starting small and then when i know its good i go bigger.

you need a pc to work with agar/lc/grain

i fore the life of me can not figure how you do it with out it. unless its the reason for the contams.

I bet you are getting contaminated because your incubation temperature is too high.  Let us know what temperature you incubate at...

Fordrew...my incubator is set at 81 F.  I thought that was the problem after the first batch of contaminated jars. I bought a thermometer to put in the incubator just to be sure.

Feelfunny...thanks for the thought. No..they arent vendors who are currently associated with this site...although each one has had a history here.

Just scrap the TiT, incubator all together.  Unless you live in a house that doesn't have heat.....

Lower the temperature down and you will be guaranteed results.  Bacillus really really likes warm temperatures ...

me too i incubate though cuz its cold here. mine is set at 78.5 deg F
