How do I maximize output from a tiny bit of spore solution?

I have about 2ml spore solution left of some very hard to find spores. I am wondering what I can do to maximize my possible success rate from these. Should I risk inoculating one bag with the last of them or should I make an LC with that little and use to inoculate more?

Go the LC. make two batches of LC with the last 2ml

Ya know I think this will be the perfect opportunity to test out my new LC jar. It's a short garlic jar, about as tall as a baby food jar but thicker around. I put a self healing injection port in the top.

I'll stick to karo/water since Ive done it before and am comfortable enough with it that I dont think Ill be as likely to mess it up. (plus grains have to be PC'd much longer) I want to make sure it takes hold.
Let me check to see if I have the procedure straight: -Add 5% volume of karo light corn syrup to boiling distilled water. -Pour into jar and seal -PC for 15 minutes at 15 PSI. -inoculate

your right about the karo, but a grain lc all you do is put some colonizing grains into the water

IMO adding the extra steps of spores > grains and grains > LC are far too risky to attempt without a glove box or a flow hood (neither of which I have.
It's something Ill experiment with in the future when I have plenty to spare and the means to do it; but I have very little, the spores are already in the syringe and I already have all the equipment to make an LC.
Im using this LC to colonize grains. If I were using the spores to colonize grains directly and had enough spores to do that without risk I would not be bothering to make the LC.
The whole goal of my doing this is to maximize output from a minimal amount with minimal risk of contamination.

I think you would need some sort of gas exchange/filter in the lid of your jar so your jar doesnt explode when PCing.
use 1 cc/ml to inoculate so you can try 2 LCs.
I personally cant get anything to grow in light karo, honey, no problem but thats me.

if you read/look at the tek its never exposed to air if you follow it as is. no need for flowhood or glovebox.

I've gotten stuff to grow in light karo just fine. I have a few cultures sitting in the fridge right now.
I used to punch holes in my lids and cover them with micropore tape. Perhaps Ill punch one in this lid and do that too.

But wouldn't pressure cooking with the grain already in it kill whatever mycellium is on the grain?

you need to read the tek,
you PC 1 grain jar
you inoculate with your spores
once jar is colonized you make a sterile water jar
add sterile water to colonized grains
extract myc water out of grain jar
let grain jar recolonize
use myc water for lotz of inoculations
repeat as needed
you can use 1cc of spores to start and still have 1cc to do your karo lc with out a problem.
just a suggestion not telling you to do it, I just wanted you to understand the grain LC its a better way to get lots of good myc cuz when you do a reg karo/honey LC you cant tell if you have contams untill you use it. wsith grain LC you see the myc is good in the grain b4 you use it.

there yu go. early admission
Pint jar on right is multispore inoculated (SA). Allowed to colonize near 100%. Syringe is 60 ML w/18 gauge needle. Quart jar on left is sterilized water.
Aspirated 60 ML sterile water into syringe.
Injected 60 ML sterile water into colonized grain jar.
Shook grain jar to liberate mycelium from grain.
Injected total of 180 ML sterile water into grain jar. (shaking mildly between each injection)
Aspirated out mycelium laden water from grain jar. (all I could aspirate out of the grain jar was 150 ML) Then, injected mycelium laden water back into sterile water jar.
Approximately 500 ML of Mycelium laden water. (enough LC for approximately 50....10 ML syringes)
The advantage of this procedure is that you can be assured. The culture in clean. Because you can VISUALLY inspect colonization in the grain jar. (if there is FUNK, you can see it )
That is not the case, with sugar type LC's. (multispore syringe inoculated) They can be contaminated, from day 1. And, you NEVER know it. Until you inoculate spawn jars with that LC. Then, have every single spawn jar GO SOUTH.
