When I mix my colonized spawn and substrate together, then place these in a tub. Do I need to do anything with the heat it recieves before I bring it into a temperature of 65-70 degrees?

Do I need to use a different temperature for the sub to colonize, before the fruiting temperature?

Not really, just keep it at the same temp your spawn was colonizing at, between 70-75.  There is room for fluctuation tho, as long as it doesn't get over 80-85, you'll be fine, colder wont really hurt, just slow things down a smidge.

What was your spawn and substrate?

substrate is straw, dung and verm
spawned with grain

Slightly lower temps will slow the mycelium cell division down a little, which is okay, it will just take a little longer.  Higher temps, like above 80-85, are primo breeding conditions for bacteria and competitor molds, so if you have to err, err on the side of lower temps than higher.

Room temp is usually fine for all stages of Cubensis, unless you live in an extreme climate, then you need to adjust conditions.  More often than not, people incubate needlessly and end up with contaminates.
