update below

So I've had several species in slants, and in the refrigerator for almost 6 years. I'm in the process of reviving them. I cooked up some agar about a week ago, and transferred some mycelium to new plates. 2 out of 10 came back. Pretty cool. just thought I'd share that with ya'll. looks like i won't get all of them back, but at least some are still viable. Here are some photos of a blue oyster pleurotus columbinus and aspen oyster(Pleurotus populinus) growing on agar.

This afternoon i did another transfer with the plate you see below. I also have started working with the other slants i have to see if they'll come back as well.








10/5/2009

just a quick update. I got 10 more plate worked up again, and i got one more species to come back to life. Its a coprinus comatus that i found in the wild back in 2004. I made a clone form two of the four mushrooms i found. looks like just the one is showing new growth. I'm happy with that thou. I'll probably grow them outdoors next year. I may follow up with pics laters.

I thought this was just the coolest thing. the pleurotus populinus is pinning on agar. heres a pic from yesterday. I looked at it today and the pins are really coming along. I'll get another pic in a day or so. I have duplicated all the plates of the cultures that came back to life, and will get them on slants soon as i can. I also want to transfer the plate that is pinning to some grain. I think i'll grow this one out indoors. I just cant wait to see her grow.:ooo: One other thing. I had a fourth culture come back to life. The morchella And i believe it's the morchella angusticeps. i inquired this one from a wild mushroom found in northern Michigan. The original plate had alot of sclerotia formed on it. We'll have to see about this one. I'm not a morel expert, but i do know that the species genetically degrade very fast. It may possibly never fruit. I'd have quite a hard time with it in my climate anyway.




This is the last update to the thread.

The photos of the plates are labeled. You can see the species that i revived after almost 6 yrs of lying dormant. I am totally impressed with the aspen oyster pinning on agar after all this time. I actually put the plate in the fruiting chamber just see what it would do. In the picture you'll see half the agar has been cut away. I made an LC with that side. I have multiple plates of all species that i'm posting pictures of, and I plan on working with them through out the winter Indoors. hopefully by spring i'll have my outdoor fruiting house done.

Cultures with plain water, or toothpick, or?  What size of slants are you using, and how many ml of the culture remained in the slants?  Thanks for sharing the pics. :sun: ~P~

well the slants were 15x100 tubes. cultures were stored with MEYA. There's not much left of the culture slants at this point. i could probably get another plate from the slants, but i won't. i have new growth to work with now.

No not all were oysters. interesting thou, the only ones that revived were oysters thou.

2 out of 10 is pretty good after all those years.  I've found oyster, shiitake and reishi can survive that long.  The others are probably toast.  Putting a piece of wood in the slant will help a lot, but five years is a long time.  I re-make all my slants once per year.

Good idea. When you talk about your farm, it makes me wonder things like; Do you have employees? Do you spend two days JUST re-starting your slants, or do you do a few here and a few there?, etc. Fascinating stuff.

I'm still waiting with anticipation to see a pic of your new grow bunker FULL of edibles! (Damned your lucky. You need to buy the wife some more flowers!)

The other eight.

cant even remember all of them,but one was a morchella isolate. The others were some cube isolates, form back in the day. Myself and a friend of mine had a thread going called The clone wars or something like that. The out come of this war was some pretty darn nice cube strains we saved. I was trying to talk him into starting it again, but i dont think it'll happen. the whole clone war thing was kinda a show down to see who grew the largest mushroom. we both had mushroom growing between 75 and 96 grams. wet weight

Shit they do look pretty big, 100 odd grams each.

Shame none of the actives worked youll have to clone some more

'grams GT #103' i interpreted as 103 g but its actually the clone label

Well The update above stating that i produced 10 more plates, was another attempt at reviving those that did not come back the first time. I wasn't to optimistic about them coming back. i'm sure it's just a loss. I also have used most of the culture media i had. It's over for those cubies. No biggie thou i have plenty to play with for now.

I suppose if you really want those cultures you can always start from the beginning, from spores or a culture off someone. Good luck with the rest of them.

ya the one that is pinning is an oyster. I put the plate in my FC now the pins are clear up over the top of the plate. Oyster's are know for pinning on agar, but after almost 6 yrs. Im amazed.
