How much liquid to put into agar plates?
    #11165521 - 10/01/09 08:24 PM (8 years, 17 days ago)

I've got petri dishes, I've got agar powder and dry malt extract. I did a prior run with ground up agar sticks into MEA, but when I put it into the petri dishes I had no idea  how much  to put in, or even if I did the MEA right. Is it supposed to be a liquid with a similiar viscosity to water? I only put enough in to coat the bottom, but it still swishes around and the mushroom pieces in there move around quite easily.

Is it supposed to be a lot more viscous, and how many mls would you put into a 90mm diameter petri dish?

I think the viscosity problem is that I tried to mix the agar into a room-temperature liquid rather than at a specific range within which it is water soluble. I've been searching for teks but haven't found anything cohesive.

Edit: Just found this:

Malt Extract Agar (3%)(MEA)(General Microbiology)(FDA M93)

    30 g Malt extract
    20 g Agar
    1 L Distilled water

Boil to dissolve ingredients. Avoid overheating, which causes softening of agar and darkening of medium color. Autoclave 15 min at 121°C. Dispense 20-25 ml into sterile 15 x 100 mm petri dishes. Final pH, 5.5 ± 0.2.

This medium is recommended as a general maintenance medium. [1]

Which makes me feel like an idiot because I did basically everything wrong haha. Although I guess my final question still stands, how viscous is the final product going to be? Obviously it still needs to be poured, but what's to stop mycelium stem etc floating around with the slightest bump?

10-12 ml is enough for one plate.

It will be more viscous than water when you dissolve it, if you weigh the agar out right the it will harden upon cooling

Always boil the water add the agar boil just for 5 sec. make sure it has all dissolved.

So the process is, boil water in the jar, add agar while boiling, mix then take off the heat. Chuck in PC, cook @ 15psi for 30 minutes, let depressurise but take out while it's still hot and pour into petri dishes, and it will solidify within the petri dishes?

Do I add mycelium when it's full solidified?

Yep pour into the plates when the agar is hot, you may wish to wait for 10 mins after you take it and pour as the agar firms slightly and will set sooner.

I wait 2-4 days before i use them so i know the plates are clean but yeah use when solid
