Q on cultivating wild samples?

Greetings,

I would like to request any advice more experienced growers can provide on this subject.  I'm looking to take samples from inner stem or mycelium growing under fruiting bodies from wild specimens and cultivate them.  From what I've read from forums here so far, it's easiest to use a nutrient agar with antibiotic mixed in to start the sample on.  Has anyone tried growing a sample on a grain substrate instead of agar?  Also, is a grain substrate like WBS (as I can't find rye berry where I live) a good general substrate for mycelium propagation before inoculating a fruiting substrate?

I have some samples, but have been storing them briefly with drop of 3% H2O2.  Is this a good way to preserve them for a little before moving to agar?

I'm taking it upon myself to collect and store (bio-prospecting) fungal samples from local woods to later test their biotechnological viability and perform digestion experiments.  I do have experience growing ordered samples, though I'm always in the learning process.  Lastly, I take very meticulous notes.  If you were curious. 

Any assistance with cultivating/domesticating wild fungus is appreciated.

To test the aptitude of certain fungal species ability to digest various substrates.  Many of substrates being tested don't easily degrade in nature.  Unfortunately I don't have access to perform chemical testing to confirm anything.. yet.  Till then, a visual comparison would have to work.

Seen seen thought so.

About the h2o2, i have seen it been done to the tissue prior to agaring it but i cant comment.

I have used antibio agar and i would definatly use it for wild samples. Dropping a sample in grain is very risky and any type of agar would be better so you can clean up the myc and be certain that your grain will will colonise. WBS if fine for spawn.

hey just a thought..

i have an idea to use grain water (whats left over after a simmer) in agar instead of dextrose, yeast, etc etc.

here is a pic of my grain water

now as you can see in the pic its starting to separate..
the separation is more pronounced now.

the idea is to use the upper part of the solution in an agar mix, thus using grain as the nutrients for the agar.

Although i may advise others in a general way regarding all types of mushroom grows, and may even post question from other forums about growing "active" mushrooms, i only grow non-"active" mushrooms and edibles.

OP, agar is the best choice for what you want to do, for lots of reasons. Most obvious: it's the only way to guarantee a clean culture.
