20 year old mushroom ground up with spores in it

it was picked wild by me 20 years ago dehydrated and put in a capsule.
it was stored under the worst conditions possable from hot to frozen cold.

if i hydrate it and plate it will it grow or is there something else i need to do?

i have micro slides etc from then when i picked it and an lc tek "new tek which was just starting out"  i printed off the net from a sight like this one. wish i knew the name of the sight from back then. well not exactly printed but notes taken from the net and mag's i was reading at the time.

i would like to pick up were i left off and try to get it to germinate.

some of the spores under the scope seem to have germinated with the squiggly things "name escapes me right now" coming out the germ pore

a high number oh 50% have no germination.

i was wondering at what rate do the spores die or what is the half life under the worst condition of spores.

this sight has helped me so much now and this is a big deal for me. i hope one and all can give some good suggestions.

i have heard of much crazier things working. i would take a small amount hydrate it and place it on some agar. if you have antibiotic media use it.

whats the worst that can happen? it doesnt germinate, oh well. i vote try it, clean it and grow it.

cool what i was thinking but i have no antibiotic but will check around

what type is best and how much do i add to my 500ml batches.

also what is the best temp to incubate at i have an incubator that is very precise and it does humidity too " egg type real big converted for my other projects"

also it is not that much so should i put a pin head amount into hydration or more.

the pin head i just put under the scope looked like 1000's of spores.

You wont be able to get snake venom and its used for something else. You can buy antibio agar easy, but it will be hard to get the antibio on its own, i think they use gentamicin Im not 100%.

you should be able to ger gentamicin at a feed store

thanks man im not so good with snakes don't you know. but i do have a good doc. so gent is the kine and what is the dose for 500ml of ag.

Oh god id use 0.1-0.2 g per 1000 ml but ive always used premade antibio agar mix.

i would take a small amount about the size of say 2mm^2. place that in a 1-3ml pcr tube and add minimal mineral salts solution, about .5ml. vortex/mix well and allow to sit overnight.

then taking a 10uL steriloop do an isolation streak on pda or similar agar. repeat on as many plate as you deem neceassary. if growth is not seen after 1 month try again with a more concentrated solution

any and all mycelium growth that is free from major contamination should be transfered to plate of pda or similar media with hydrogen peroxide. from there just treat it like anyother muchroom culture
