Total Time of (PDYA) agar preparation

I am trying to figure out some good times in executing the steps in agar production. For this thread in particular it will be PDYA but others may ensue
Your opinions are welcome!

(1/2 liter solution prep)

These 2 steps done together...
Boil a couple small potatoes in 2 cups water. (30 min)
Boil yeast nutrient to kill it (30 min)

Combine agar flakes, warm potato water, yeast nutrient and honey in a clean bowl STIR (5 min)

Put into flask/container for PCing (1 min)

Heat up PC to 15 psi (10-20min)

PC (45 min)

Let PC cool so flask is able to be handled bare handed (90 min)

Pour Plates (gets faster with experience)

total time:
3 and half hours give or take some time

i just started using professional agar and malt extract
if you have the money buy that
takes like 2 mins to measure out the amount to use
mix that
pc the bottle with plenty of water in the pc to avoid boil over, which is a real
pain in the ass
heating up the pc takes about 20 mins then another 40 to pc the stuff
then let that cool in the pc for a cpl hours and remove it so that its still liquid but barely cool enough to pick up, pour it and let that harden in the petri dishes
probably like a 6 hour process till you can put spores on it
maybe even more depending on cooling times and the agar solidifying in the dishes

Stamets says to do 45. I've done 30 a lot and likely heard the figure of 20 as well.

sillybilly, i like using potatoes and honey cause i can get them from local farmers, but thanks for the suggestion I'll keep it in mind.

I use 15 min at 15psi and get good results. That's what we always used in lab, so I just kept doing it.

Note, this works better for thin glass containers and things, you really need it to heat all the way through. I use a 500ml florence flask for most of my agar pouring. Works wonders.

Ya know if your using honey, it may be a good idea to pc it for less time to prevent caramelization. I'm not sure if dextrose caramelizes at the same rate but I'm inclined to think honey caramelizes faster.

If your not familiar with caramelization it is the breakdown of sugars of which honey is supposed to provide for your purposes.

when PCing a flask full of agar concoction can it be sealed off or should it not be rubber sealed for so pressure doesn't build up

I use double tyvek with a rubber band, then foil over that. Never had an issue.

I would stay away from stoppers.

how long is recommended to let a flask cool in 2 inches of water IN a PC

it varies, but you can take it out as soon as pressure is neutralized so long as you don't drop it cause it may still be hot.
