how long do syringes stay sterile?
    #11065476 - 09/15/09 08:08 PM (11 years, 7 months ago)

i sterilized some syringes in a pc as follows:

- rinsed
- filled with 1/2 distilled water, 1/2 air
- wrapped in foil
- pc 60 min. on top of WBS jars

i stored them for a while in the foil, in a zip lock freezer bag and did not open until it was in the glove box to fill with spore solution. now when i say i stored them for 'a while' it was a few months. i felt at the time that if the water left in it was distilled and in the pc for 60min. it would be O.K. after using said syringes for 4 strains in WBS i achieved 100% FAILURE. however all the prints used to make the syringes were questionable.

so i guess my question is, did i totally screw the pooch by storing the pc'd syringes so long?

also, how long can they sit after pc'ing?

me too, they stay sterile so long as they don't come into contact with anything and have been sterilized and placed in a container to prevent that.  If you leave them just lying around, you may have introduced a contamn.

they were in a ziplock in a cabinet i keep all my other sterile stuff in. i just would have thought i would have got at least one good jar.

i think i fucked up the sterilizing some how. they contamned same way, same rate.

i put some drops from each syringe in agar so we'll see what happens but i think i'm gonna trash the rest.

To sterilize my syringes all I do is boil water for 30 min while thats do that I suck up some 99% iso in my syringe and swirl it all around then after 30 min I expel the iso into the syringe cap while its prop up. Then I suck back boiling water let it sit for 1 minute then expel and do that 5 times then on the last one I keep the water in it then take my syringe cap shake the iso out of it and cap it and bag it. Always works
