Grain to agar..........

I was just curious if anyone had any luck with taking a piece of grain that has been colonized in your jars and placing it on a petri dish of agar. I always seem to have some that stick to the glass and never get introduced to the bulk. So would I be able to grow out the kernel of grain and get a new culture to place in more grain later? Just a thought, if not, I have plenty of spores.

Just put the grains in more grain. Skip the agar unless you are going isolation.

I think he wanted to use a grain that had already been exposed to unsterile conditions tho. If he tossed it in with fresh grain it would probably contam. If he goes to agar he can separate out a clean sample.

I do all my work, even setting up monotubs in front of my flow hood, so it should still be good, but want to do some isolating on the grain that is left over. I have a good strain of TC and want to put some in storage.

G2G is relatively effective for isolation. You don't get a choice as to which strain takes over, but that probably doesn't matter as a very strong strain will usually be dominant.
I have been very successful with always rolling over one jar from each initial inoculation to spawn new jars. You can keep this up for several generations.

i just made 15 h2o2 tek plates and put some grain from the jar i opened for use on them. this is my 2nd try. the 1st i put too much h2o2 in the auger and the myco stayed confined to the sample. now with less h2o2 it should take off. in this test i have shittaki and portalini clone tissue from the g-store too. so its an all around test on the h2o2 plate. i also did it with no gloves and no hood in open air.
are you going to try it? it would be nice to hear someone else doing the same thing just a little different.
edit add--- i also put red food color in the aug to help under the scope. might try mixing colors next time lol
