I'm working on documenting my grow techniques from start to finish, this is the first part. Suggestions are welcome, and if anything doesn't seem clear, please point it out, I can revise.

Directions for creating a liquid culture jar, with gas exchange and needle port, for use in a pressure cooker. Using the vacuum tube and sealant is something I learned from agar. These are modified with home made sealant ports. Each lid will hold up to repeated use, though they will wear out eventually. Make them in twos for redundancy.

Parts:
Jar -w- lid and rim
Drill -w- 3/8" and 1/4" drill bits.
3/8" automotive vacuum tubing
RTV high-temp gasket sealant
Polyfil



Parts Notes: I'm using pint jars, but any sized jar will work. Small mouth will work, but wide mouth is preferred. I'm using wood bits, as they seem to do a better job of making nice holes, especially at 3/8". Bad for the bit obviously, but it will last through quite a few lids. Just the right amount of pressure at high speed will keep the metal from tearing. The vacuum tube and the sealant can both be found at any auto supply store. I've always used RTV sealant, though I've read that other silicones will work. Polyfil is a synthetic fiber used in pillows and other stuff, and can be found at various places including the 'marts.

In this next picture, you can see where to drill the lids (done while lids are secured to jars). One 1/4" hole and one 3/8" hole, opposite each other, both around 3/4" from the edge of the lid. Cut a 2" piece of vacuum tubing for each lid and push them about 3/8" through the top of the lid. Should be a nice tight fit.



Apply sealant around both the top and bottom of the tubing, making sure that the tube and lid have coverage all the way around. Apply a blob of sealant over the 1/4" hole. Flatten it a bit and turn it over. There will be a small amount of sealant poking through the hole. If you push it back up, this will cause a nipple to form on the top, which is good for aim.



Flattening the sealant is best done with some spit and a finger. I've also had some luck with a small piece of wood (popsicle stick, clothespin) which has been soaked in water for several hours, but spit gets the job done better IMO. Once the needle port looks good, set the lids aside in a well ventilated area to dry for a day or two.

After they are dry, place a small piece of polyfil in the tip of the tube. If you take a small piece, twist it, and fold in half, you have a nice tip to push through the tube. A toothpick can be useful. You don't want the polyfil super tight, but you want it tight enough to keep contams out. With a little practice you can come up with something reasonable. Trim off any excess fiber, and these liquid culture jars are ready to use.

Nice tutorial, can you shake those to loosen the mycelium without getting the polyfil wet?
