Golden Teacher

alright so right now I made two WBS quart jars but I don't have a PC so I figured I would just trash the WBS,


but anyways when scraping spores into a shotglass I put too much water and so instead of wasting it i figured i would just poor it into the WBS jars, which i did completely unsanitary since you guys said i had an almost sure contam rate anyways,

but here we are 10 days in and the first jar shows nothing but the second has about a 2 inch wide white spot showing up on the side of the jar, and no contams

BRF JARS

6 days ago i inoculated 3 quart jars with 1cc of hydrated spores

so far i only have one jar with some small but noticeable growth, is this a bad sign? maybe i just didn't give them enough spores but i wanted to clarify, THIS IS EXCITING!

ill keep you guys updated, im going to upload some pics of the ones with growth tomorow

Good luck, but getting mycelium to grow in the jars is the easy part.. People like to see photos alittle later on I think =D. But awesome!

you should always keep up sterile procedures. That said, poor prints are a bad move, if that's what you have. Printing is harder than you think, they should be dehydrated with desiccant for long term storage to prevent contamination. I had a print that I scraped to agar and every single dish from it had bacterial or mold contaminants. I may try it again but that seems like a lot of shit that something like 5 or 6 dishes contaminated that quickly. Seems like a waste of time to me on that print, while another similar amateurishly stored print made quite a few uncontaminated dishes I cultured out.

these prints were from a sponsor so im pretty sure they aren't contaminated, do you think its just taking longer than normal?

10 days is a long time, particularly for a sponsor. Are you sure you didn't do anything that would kill the spores?

well i scraped them with the needle of the syringe into a shot glass and it kept powdering across the whole shotglass and getting a little bit on the counter beside it, so i think it was a lack of spores,and the WBS were the ones i inoculated 10 days ago, and one of them is showing signs of colonization, but the other had a visible amount of water collected at the bottom and no signs of colonization so i think it might be a failure, what do you think?

well just a few things ive learned off of here, not growin yet :smile:

1) try to find something else to scrape spores with, like an innoculating loop...that needle could kill some spores easy.

2) well if one is showing then you atleast did something right, congrats! :smile:

3) visible water is no good in jars...make sure its field capacity next time!

i say just give it time, worse case atleast u got one goin!

Well I'm inclined to think you should at least see bacterial growth by now if it were a mater of the print quality. Is it possible that you did not shake the needle enough to get any spores into the first cake?

i didn't shake the needle at all :/ so how long should i wait before i ditch it? i dont want to open it up and see it colonizing and have to throw it out :frown:

alright so now it is 6 days after inoculated the verm and on one jar there is 3 inch spot where mycel is growing and on the 2nd one there is a 1 centimeter spot and on the 3rd there is no sign of growth AT ALL helppp

ok I see. Give it 3 more days on the BRF cake. If no growth is present, make a new one. Spores can take a while to germinate sometimes 5+ days. After 12 days if no growth is present it's probibly not worth it to keep it longer.

Personally I'd toss whatever you have of those WBS jars. I've done "fractal(steam) sterilization" on grain jars before and they have less than 50% success rate and there is also an added danger that they will be contaminated but you will not be able to see it on the outside of the jar. I had lucked out and correctly identified the contaminated jars I had, but it is harder than you think to do this properly and can take a hella long time to identify properly. If you wish to keep your WBS jar watch it like a hawk for separate contaminant sectors or any greening.

3- ive had more success with a little extra water in popcorn jars, then with field capacity with RR's rye tek, i figure if you could grow mycelium in lc's which is what, sugared water, why cant it grow on grains with a little extra moisture,just spread the moisture around all the grains, i dont get it.  i figure its much better to have a little more water than just enough or barely.
