isn't this supposed to be a standard lab procedure? I haven't bothered reading on this but from what I've observed of lab procedures for growing out bacterial cultures, you swab left to right (or vice versa). And with each stroke gradually move from front to back (or vice versa). Making S like curves across the whole plate.

But this is how I would do it.
and I forgot to add this to the post, rotating the swab also helps to grab things up, but when swabbing I would roll it back and fourth so that areas with spores would rub on the agar. (I wouldn't roll spores on the whole surface of the swab, just the bottom half)...
*this should be interesting if someone corrects me, I'm gunna need to know this too and would like to see if my assumptions are good on this*

I don't think you should dry the stuff that just sounds dumb to me, myc needs moisture.
The agar should probably be a gel-like consistency.

well, how hard are we talking for these agar plates? should be soft, yet firm...with a slight bit of moisture on it possibly.
swabbing is done in the manner of which xtofury said. depending on technique, and item used for swab, you might not *and preferentially not* ever touch the agar with the swab, you inoculate by simply dusting with spores. now if all you have is a q-tip, and no spores readily fall off, then sure...touch the plate. but know that touching it is only going to possibly introduce contaminates.
but point remains, back and forth, kind of a Z like pattern, as to inoculate a bigger area, and create a better genetic mix.
hell if you want, just inoculate 2-4 different points (rather than streaks) in different quadrants on the plate, that way you can pick the best of the 4, selecting rhizomorphic growth. with the Z pattern, it becomes harder to isolate because of overlapping. still possible though.

i just tried swabbing , nothing happened..........
i dont understand how syringes need to be left 24 or 12 hours to hydrate from print, but we can swab dry spores onto a jelly like substance...
i need a better technique also. not had time to try again.
