is this wbs pint ready to be used to make a grain LC?

i feel like i post on this site way too much. I'm sorry to all you people that are getting tired of my questions... :frown:

anyway i have this WBS pint jar. its about 1/3 full of wbs. i inoculated it with spore scraped from a spore print on 7-26-09. today (8-5-09, 10 days since inoculation) it looks like the jar is almost done colonizing. I've heard that when making grain liquid cultures many people make them before 100% colonization.





what do you think? should i get my water and syringe ready to spray water in and shake etc to make the grain LC?

if its not ready, why? and what will it look like when ready? don't get me wrong I've searched and found plenty of pictures etc. i just want to make sure

I'm not sure about making the lc before it's all done. I've never heard that.
If it's 100% done and no visible contams or signs of distress, shoot it up.
I say go for it!

Just wait for around 95% colonization and then go for it!

Also, after you're done making your LC, put your grain jar back into incubation.  The mycelium should recover and you won't have to waste a jar of grain.

Because it would work?  Anywhere between 95% and 100% is fine.  The more mycelium on the grain, the more mycelium you're going to get in your LC.

So you'd obviously want as much mycelium as possible.  95-100% colonization is fine.  You could do it when it was 50% colonized and it'd probably still work, you just wouldn't have as much, so you may as well wait until it's almost colonized (95% minimum, in my opinion).

Until it is done, you may have hidden contams. (Still after then, but that's another story.) Growing a bunch of mold quickly doesn't get you anywhere. Slow down. Wait for 100% colonization, then wait a week for the mycelium to really get a strong foothold.
Sure, that's what they say about cakes, but I do it for Grain LC for the same reason, and I recommend that everybody else does too.
LC is hard enough if you do it well, there's no reason to half-ass it.

I'm trying to copy what you did on your mini mono tub so if you say wait, i guess ill wait. :frown:

at the moment I'm guessing its about 90% colonized (its been a while since I've grown..) so ill try to slow down and at least give it a few more days.

here is a jar i just finished PC'ing, 30 minutes. its 150ml of spring water (store bought) in a pint jar with an rtv injection port and whatman 0.2 um filter. unfortunaltey the filter did not do so good in the pressure cooker. i guess too long?



oh and project, Ive read about reusing the grain jar. isn't that amazing how well it comes back? i haven't seen the maximum number of reuses but i bet i can do at least 3 safely. why do people still use annoying LC?

Wait until 100% shake and the next day if it looks like it is recovering then use it for a grain LC.  If it doesn't recover don't use it,  you will just contaminate more jars.

one more question.

i know what everyone will say but i just need to ask...

what if i used a very low concentration (0.1%? lower?) of 3% h202 added to the sterile water before injection into the jar? i know many people say never use the stuff but it would make me a little more comfortable.

yay or nay?

Never use the stuff.

Wait at least until 100%. If you can't wait longer, then so be it. :smile:

And I don't know what you did to that filter, but it's dead. And what's the filter for? You don't need a breather port.

it melted. it was brand new and sterile in the package. i wanted the port so that when i sucked out the water and injected the myc water i didn't want to expose the jar to unclean air and or produce a vacuum. its the way agar does it on his grain LC post.

i think it will still serve its purpose, its just hideous :frown:

OK i officially have no patience at all! i knew i would end up just doing it no matter what y'all said. i have failed you.  :sorry:

however the mcy water looks amazing! i did not dilute it but i have 16 gauge needles so there has been no problem with the viscosity. do it anyway? eh i dunno i hate risking contamination. my largest needle is only 30ml.

a lot of water is still in there, maybe 20ml. so ill keep rotating as often as possible.

here is a picture of the grain LC and the grain jar used . i shot 10ml of the grain lc into the bag. i know this is higher than you all recommended but i made the spawn sorta dry, which is why i added the verm, to suck up excess water. im paranoid about contams and never adding enough. i suck. :frown:



i know no patience makes this hardly the perfect hobby for me, but when success finally comes again i will be able to slow down. will update as soon as i see growth. im pcing 2 pints and several bags of wbs right now.

Get the extra 20 mL out as fast as you can.
I never get as much water back as I put in- how did you get so much?

i got 20ml less out than i put in, that's whats left in the jar. i just tipped the jar so that the lid was facing a 45 degree angle below the horizon. then i stuck the needle in and all the water pooled down right at the injection port. but that 20 ish ml left just didn't leave the grain. its still stuck on it and is quite hard to get out. agar had the same problem when he posted this tek. ill try to grab some more but it ain't easy. :wink:

i don't know evildee, i never saw the smiling one but i do like the current one. its unique :wink:

but to litcloset.. y not wait till you have fruits and then make an lc/clone... ive read before about what your doing but dont know why its done.. ive only done it when i have but one jar of a species/strain i dont wanna lose..

the reason i did it was to quickly colonize the spawn bag that i have. its done instead of g2g because it allows working with the media while never opening it to potential contams. it also has been claimed to colonize quicker because of the many strands of myc compared to individual grains.

also i can store it in the refrigerator for a long time and use it to inoculate many more than 10 jars from one jar unlike gtg.

plus i failed at LC this time around so i had to cheat
