birdseed probs
    #1062875 - 11/18/02 02:05 PM (21 years, 4 months ago)

Okay, this just confuses me. I drew a 20cc syringe of well colonized Karo water. I then proceeded to inject three birdseed jars with 6cc's each of this solution. I shook between each inoculation, and this is what happened. All that growth has happened in about 2 days, three days ago they all looked the same.

These have all been incubated at 83-87 degrees, no light, inoculated 24h after PC'ing at 15psi for 50min. The only thing I can figure is that the mycelium all clumped up, and then shot into one jar, and I just shot water laced with trace amounts of mycelium and corn syrup into the other two. Any other ideas?

i really have no idea what you are talking about. i can try to piece this together..... let me try....
I drew a 20cc syringe of well colonized Karo water
no idea what that is all about.....

I then proceeded to inject three birdseed jars with 6cc's each of this solution
im guessing there were spores in this "solution"- way too much. you only need 1 CC per jar.

The only thing I can figure is that the mycelium all clumped up, and then shot into one jar...
hmm....maybe you are talking about spores clumping up in the syringe, and all went into one jar, if so, you should remember to shake the syringe up a lot.

...and I just shot water laced with trace amounts of mycelium and corn syrup into the other two
is karo water=corn syrup? maybe...did you make these syringes yourself??

i hope i helped, but i dont know what to even answer.

I think he made a liquid culture, used it in syringe form, innoculated some jars and only 1 came out with mycellium.Thats What I got out of it atleast

OK, Karo water = a liquid culture in which spores are germinated, made of 100ml distilled water to one teaspoon of light corn syrup
Not spores, there were no spores involved in this! The mycelium, growing in the liquid culture. I was thinking that in the syringe, the mycelium clumped up inside the syringe, meaning that in the first jar that I inoculated, all or most of the mycelium in that syringe went into that jar, leaving just corn syrup + water in the syringe, which I proceeded to inoculate the other two with.
Yes, these were home-made karo water syringes.

For some strange reason liquid cultures have the lowest % of success out of every other mycelium tissue transfer tek.Lets say, hypothetically, out of every 12 jars 3-5 make it.Now multiply that by 20.Not a good average. So why keep trying? The liquid is the fastest way.G2G transfers work almost 100%,but are slower.I first thought that to much h2o2 could burn the mycelium, lower the h2o2%?No change. How about no h2o2?No change. The jars never started. How about a G2G transfer to one of the jars that did'nt start? It worked. So it's not contamination.I think putting a fat spoonfull of mycelium from a nice jar into clean water and shaking it works better then growing in liquid, but don't stop trying.

That is precisely what happened. I have used the Karo tek extensively and unless I am extremely careful, the first couple of jars take off and the last one or two grow slowly if at all.

Mspacman I am sorry but stick to PF-tek and read some more. I am not sure why you are having problems with your birdseed man. Give it a few days.

When I used liqued mycelium only 1 jar turned out good and I was thinking it was because of the same thing and now I am pretty sure thats what caused it.

I believe the trick is to make sure the mycelium in the karo water is well broken up into little pieces instead of several big ones before you suck it up into the syringe. Then you want to shake your syringe up real good before you innoculate each jar. I often shoot up to 10 cc into my birdseed qt jars because I have lots of mycelium in my karo water and usually need to use it up before it dies. ( I cannot put it in the family fridge for storage.)

schmoppy if you start to see contams then try shooting for 90 minutes in the pc when using grains and birdseed.
