just take the syringe and shake it up. use it and if your outcome isn't what was expected, then you know where to point the blame at.

Yeah, it's fine for makinf a spore syringe, but in this case the spore syringe has been ready made, so sucking in air, even "sterile air", is just an unneccesary step and therefore introduces a greater chance of contamination.

In short, give the syringe a shake :laugh:

im not trying to call you out or flame you. i was telling him my opinion on what to do, and in my opinion, sucking air into (an already made) syringe is not a good idea, especially burning hot air from a lighter.

and aside from knowing information before you advise someone on it, you should read the post word for word before you try to prove a point. no one was talking about making syringes.

That?s not true.
The hot, sterile air, immediately cools down when it comes in the contact with the water in the spore syringe and the temperature of the syringe rises virtually nothing, sicne the heat capacity of air is so much less than that of water.

As for shaking a completely full syringe, you can shake it for hours, and it still won?t be shaken properly, since the fluid in the syringe can?t move properly.
That?s why it?s better to have a small air bubble in the syringe.

As for "if you suck air in the syringe you will contaminate it"; If you suck in the hot air, you won?t contaminate it, 100% guaranteed.
Even if you suck in "normal" air, the chances are still slim the syringe will contaminate because of this.

Try it. Take 2 identical spore syringes, suck 1 ml of air in the one, and leave the
other completely spore filled. Shake and observe.

I said "suck sterile air heated by a flame" into the syringe.

And I said that even if you weren?t sucking in sterile air, the chances you contaminate the syringe are slim. I?m saying so because I tried it on several occasions.

Because the spores woudn?t shake up otherwise, I didn?t know about the sterile air over the flame at that time (a few years ago)

The contaminant load of the air we breath is slightly overestimated.
I don?t advise it though neither , if the spores are hard to shake up, I advise to soak a bubble of sterile air from above a flame, it?s easy and guaranteed contaminant free, why take even the smallest risk, if there is a better way of doing it.

Again, try it, and see for yourself.

I totally agree with Anno on this one.

I've made hundreds of spore syringes and never screwed so much as one up pulling air into the syringe through a flame.

Try it kids! It's the latest craze!  :grin:

Everytime I don't suck air into my syringes I get clogs... then I get pissed off that I have a clog and shoot off 5 ml's into one innoculation point...

Even though I generally shake my syringes out of habit, I've found that it really doesn't make all that much difference... Sure on occassion you will get one or two inoculation points that will take off faster than the others, but in the end the cakes usually finish in a close tie... Also, most of the syringes from skrowerops usually have a small air bubble in it. If not, rather than disobeying your mother and playing with lighters, try sticking it into the jar, squirt your first inoculation point, find an air pocket in the jar, and suck the 1/4ml of air into the syringe... If you done did things right, it's pretty freakin sterile in there... and if you suck up contams into your syringe from within the jar, my friend you are in for some disappointment... this to me is the easiest, least time consuming way to do it, and I don't have to worry about burning my fingers off with those dangerous lighters!

i have only inccubated jars twice in my life but both times i have put the needle to flame to suck in a tiny air bubble.. There is no way to shake that syringe poperly without one. Both times most of my jars have colonized properly.
