It isnt rocket science either. Why the hell did you think you crossed cubes anonojon? You think you got some magic special powers or something that you can add two iso spawn jars together and BOOM you have crossed cubes? Its pretty obv IMHO wat happened

it was just 2 isolates tho that looked very distinct from one another and came in waves of one phenotype or the other. u still think both strains were present in the fruitbody?
only because RR has told me that b4 and he has been under the scope alot more than I have.
if it were 2 diff species they would colonize as much as they could and have fruiting zones of sort. one on one side and the other on the other side. like this.

it was an accident and you are a douche. when did i say they crossed? i only said what happened and what a TC told me. Read...Think...React, in that order.
RR get misquoted about the multiple strains existing in a single fruit all the time.
He said it "CAN" happen. Not that it happens all the time. 90% of the time if you take a very small tissue sample and drop it on agar, you will get no sectoring whatsever, a single isolated strain.
It's like the difference between saying:
A. A person can be born with their sibling embryo embedded in their body.
B. People are born with their sibling embryos embedded in their body.

Bah, clearly i'm not being clear.
I didn't get a cross I got a flush of 1, then the other. I never tried growing out the spores, because I DOUBTED IT WAS TRUE THAT THEY WOULD MIX.

Mixing spores of 2 strains in a single syringe, inoculating agar, doing transfers and then fruiting could possibly give out a strain combination, if the monocaryotic(did I spell it right?) mycelium of one strain combined with the other, this however is only a theory.

The pros can identify and isolate monokaryotic bits and put them together so they can try to mate.
There's also a way to stress a dikaryotic strand to divorce back into its parent strands. I think it was fastfred who said that, but didn't elaborate.

Yes, but I was wondering if the method I described would work because you wouldn't need any special equipment or training other than managing agar.

I wonder if there's any way to visually see if a sector is monokaryotic or not. Like a little more wispy and less thick perhaps? What if you put a single drop of spore solution on agar in a few different spots, and as soon as you see germination, took a sliver at the end of one of the spots? Could that work to get a monokaryote?

True, but with a microscope comes a flow hood, because how are you going to isolate a single spore in a solution under a microscope in a sterile fashion with a glove box? Or look at a plate under a microscope without contaminating the plate, unless you have a way to use the microscope in a sterile environment, which is impossible for a glove box.

usb cam attachment , to a laptop ontop of the glove box
i could easily do this but its not x-mas so i dont have my microscope.
dont give advice unless your 100% on it please

Where in the past 10 posts that I've made on this subject was I giving advice in any way? I'm just trying to better my understanding of the subject.
The cam attachment is a great idea! Didn't even think of that.

im not saying using the cam will be easy , i bet it would be hard, but better then buying a flow hood i bet it would be pretty hard, i guess thats where a good monitor comes in handy,
mixing strains isent hard to understand
you just need to get the terms right,
strain is -cubes, pans, and rieshi
substrain is- the sectors in your petri , so like. potent isolation of b+. good pinnset clone of b+, even though its a clone its a substrain
races, or types or verities is like , B+. pan jamica, elixir azure, azure Germany
other then that , its pre basic

you just need to get the terms right,
strain species is -cubes, pans, and rieshi
strain is- GT, Amazon, PE, Oak Ridge, and so on.
substrain is- the sectors in your petri , so like. potent isolation of b+. good pinnset clone of b+, even though its a clone its a substrain
races is another term for strain
other then that , its pre basic
