Tried to grow spores on agar. What are all these?
    #10231322 - 04/25/09 01:43 PM (14 years, 10 months ago)

Hey,

I've been experimenting with spore prints of golden oyster and shiitake, just streaking plates of PDYA with the spores under a glovebox.
Here's the sort of growth I got after 10 days. The various patches of white all appeared around the 7th day, not really matching my strike pattern at all.

Shiitake plate:

In the center, quite cottonous growth like I get from my oysters. Is there a chance the grayest of the spots, or the right, would in fact be shiitake?

Golden oyster plate:

Ugly orange bacterial contam on the edge. Are the others mold or mushroom mycelium? Anything that looks like golden oyster and would be worth transferring?

Looks good. (Except for the blotch of bacteria, of course). The rest looks just like what you'd expect.

I'm now pretty sure there's no mold since everything's still pure white.
I'm quite unsure all the patches of mycelium are the same species though, they look quite different and some started making clear demarcation lines where they met.
I guess I'll just try making grain spawn from a few of them, and fruit if it smells oyster-like.

They are all the same mycelium. The difference in their looks is genetical. This was a multispore inoculation after all. Transfer the clean mycelium away from that bacteria soon to give it no chance to grow over your mycelium.

They seem to be more or less the same, I agree, and what worries me most is that one plate was supposed to be shiitake and the other an oyster. Do I have any shiitake on the shiitake plate?

RR posted pictures of 2 shiitake strains in this thread: http://www.shroomery.org/forums/showflat.php/Number/7843769#7843769
What I have here doesn't look like that at all...

The shiitake is a 'maybe', but the oyster plate looks more like mold than oyster mycelium.

So it's been a week since the original pictures. I've been a bit lazy and didn't try transferring anything yet.
The plates are now fully colonized, and still completely white. Does that rule out the mold hypothesis?

Thanks RR. I'll just trash them. I wouldn't mind growing surprise mushrooms but not molds.
