A few tips to add to Agar's tek if you're doing this for the first time(which i started from):
Whatever jar you use for the grain culture, make sure you don't fill it past 1/2 full(as his pictures indicate). It makes for difficult shaking :B
You can (and i always do) shake the jar beforehand and break up all the kernals. I'm not a fan of vigorously shaking jars that have a hole in them and water sloshing all over. If it's already broken up before you start, you don't have to shake half as hard. (this also helps alot if your grain if fully colonized)
If you don't have rubber stoppers, high temp RTV silicone is good to use as a self healing injector port.
If you're using a glovebox and don't have experience with 60cc+ syringes, practice it in your glovebox with just water in jars beforehand.
If you plan on storing the suspension, a magnetic stir bar and plate are a good idea to quickly and efficiently mix it back up after it's been sitting a while.

I didn't read that tek yet, but is what i do is once i have a fast growing grain jar, i have to make another hole in the lid closer tot he edge, so i sterilize a nail with alcohol, and make another hole (In the Outside Air). Then, i inject sterilized honey water into my jar sideways. If i can reach the mycelium i give it a stab with the needle tip to knock off myc. then i just tilt it back and suck it up. It's usually totally white and murkey.
Inject that to get massive growths.

I'd recommend following Agar's tek. There's no need to (and i never would, personally) inject sugar water into an already colonized jar. Use sterile water and shake the mycelium off of the grains.
Adding sugar water to the mix just give contaminates something to grow on. If you extract just mycelium and water, you have a much slimmer chance of contracting contams and it will increase the shelf life of your liquid suspension.

My thinking is that if it's just myc. and water in order to grow it out you're gonna have to add nutrients.
You could suck up the myc. water, THEN add it to the nutrient liquid mother jar to multiply.
For the record though, no contams, yet.
