Revive grow with petri dishes

Hi all,

Trying to restart a grow, hoping you can help.

I have some mycelium from my last grow of some very successful ecuadorians. I'd like to revive this strain, but the dried matter is over a year old and clearly not sterile.

What procedure would sterilize the mycelium while still allowing it to grow? I was thinking soaking in alcohol would be effective.

I also have a syringe of a strain of something (I've forgotten what). Can I inoculate a dish with this as well? Will that be successful?

It might die if you put it in alcohol. Maybe just rehydrate it with sterile water then throw it on agar, then isolate healthy growth away from contams

You can wash the tissue in a jar of sterile water, then transfer it to a second jar of water with a scalpel or syringe needle. Then you wait until it begins to revive and you can transfer it to agar to start isolating away from contaminants.

So, I pressure cooked some water in a mason jar and placed a piece from near the root and some stem as well inside while it was still hot.

Hopefully I didn't kill anything by putting it in the hot water, but the idea was to sterilize. The petri dishes are cooling tonight and I'll be putting the culture in when it's ready.

How do I know when it's ready, though? Will it start growing in the water? I put a little agar in there as well in hopes of helping it out. Hopefully this will work out.

How hot was the water and how long has the mycelium been in there? Short exposure (under a minute) to hot stuff like 140-160ish and it will live, but for any length of time, like over 10 minutes or so and it could be dead.

The tissue doesn't need to be sterilized, because you can isolated it from the contaminants on the agar dishes.

It's probably ready. All it needs is a dip to rehydrate a little tissue.
It probably wont start growing in the water

Took it out of the cooker (had been in for 20mins) and put the stem in and left it in there.

It has some white hairs extending from it. Going to throw it in a dish and see how it goes.
