Acquired some corn syrup. Had a choice of a few. Chose light corn syrup w/ vanilla. Didn't want the vanilla in it, but than, didn't want dark thick corn syrup, purely for contaminate visibility reasons. Was I wrong to get the clear "light" syrup w/ added vanilla? Should I just acquire the basic dark corn syrup?
Here is the recipe: Potato Dextrose (Yeast-extract) Agar (PD(Y)A) broth from boiling 150 grams sliced potatoes in 500ml water for 30 minutes(add water to 500ml) (You can use 5 g of instant potato flakes instead) 9 g agar agar 7 grams dextrose( = glucose, or 10 ml honey or corn syrup ) 1 gram brewers yeast or yeast-extract (optional)

I have never tried using corn syrup with mine.
I do use PDYA and MEA.
I follow pretty much the same recipies as are stated in the FAQ.
Copy and Pasted from the FAQ :
Quote:
These recipes are designed for agar maintenance of Psilocybe species. All media have a near-neutral pH and should be sterilized for twenty to thirty minutes at 15 psi pressure. 500 ml of medium will pour around 20 petri dishes. Using a different medium from time to time will prevent the culture getting used to one medium.
Malt Extract Agar (MEA) 10 grams light malt extract 9 grams agar agar 500 ml potable or distilled water
Potato Dextrose (Yeast-extract) Agar (PD(Y)A) broth from boiling 150 grams sliced potatoes in 500ml water for 30 minutes(add water to 500ml) (You can use 5 g of instant potato flakes instead) 9 g agar agar 7 grams dextrose( = glucose, or 10 ml honey or corn syrup ) 1 gram brewers yeast or yeast-extract (optional)
If you read the PDYA one there are several different ways you can make it.
This is the one I make.
-PDYA- 500ml water 5 grams of instant potato flakes(Potato Buds Brand) 9 grams agar agar 10 ml honey (Raw natural pure honey - Beemaid Brand) 1 gram of Nutritional Yeast
I switch back and forth with the above PDYA recipy and the MEA. And have always gotten great results.
Click here for all the agar recipies that are listed!~
good luck!~

Be sure your pH isn't too much acid or basic.
Be sure your medium into the petri isn't full of water drops.
Be sure your specimen isn't old.
Be sure your oyster mushroom isn't a commercial mushroom. I'm not saying you can't clone commercial specimen, but a lot of time they come from a strain so much expanded that they neither haven't the power to grow a bit.
Try with others oyster first!

Might want to mention that none of these recipies produce PDA they are all modified PDA recipies.

Can you use 1 gram of regular yeast instead of brewer's yeast, yeast extract, or nutritional yeast?
Is there a difference between the yeasts?

the regular yeast will work but the others are more suited to the task and typically far cheaper, brewers and nutritional yeast can be found at health food stores and often in drug stores
BTW... the thread is a couple of years old

If this thread is that old I might as well ask my question here. I am trying to do agar work. I have made some agar using the recipe RoadKill posted in this thread. The agar that I make only gels up when I put it in the fridge. I don't know where I got the idea that agar should turn to gel, but I'm under the impression you want the agar to gel up. Should it be in a gel state to work with? I can't find any info on it.

Yes, it should be firm at room temperature.
You may need to add more agar powder.
Good luck,
