Yeah, but wont the syringe become unsterile while the plunger is removed, and then the plunger will definitly not be sterile while allowing spores to fall on it...

Correct?

Not if you open everything in front of a hood, then close everything back up to sit long enough for printing.  Then, open again in front of the hood to hydrate the spores.  Cap, label and store.  It's a good idea in my book.

You might just try printing (sterile- or as close as it ever gets), on foil, opening the syringe in a sterile field and dropping a small piece of the print into the syringe, the add water, shake, cap label, etc.  (Technique is called backloading  a syringe, btw.)

I am with them on the sterility problem. You can't wait for spores to drop straight down in front of a flowhood, it just won't happen (not to mention running the hood for hours). If not in front of a flowhood, you still need good FAE in your FC so you may have spores (bad ones) coming in that way.

The only way I see this working is in a monotub-like setup where the holes are filled with polyfil and the lid stays on all the time.

You don't need to wait for the spores to drop. Just rub the plunger on the gills like a swab.

It's used in the pharmaceutical field all the time.  This is a perfect example of why newbs can't ever search something out to find it.  Googled "backloading syringe", last link of the first page of results.

Good point. Don't we generally assume all mushroom fruits to not be sterile because of the FAE that comes with fruiting conditions?
Isn't this why we use swabs on agar, rather than going straight to grain or putting the swabs in water and using it as a syringe?

Hm, seems to me that when a mushroom sporulates, the spores have not been exposed to anything and we use that to our advantage to get clean prints. The clean prints are then used to make syringes.

Unless you mean that vendors somehow use swabs or this backloading method to make syringes?

You just said we assume all fruits aren't sterile. Vendors make their syringes straight from prints as far as I know. Those prints are sterile unless proper precautions weren't taken.

The fruits aren't, but the spores that are between gills are. When you use a swab and touch the gills, you have a pretty good chance of picking up contams with it, when the spores drop, they have a very good chance of being sterile.

At this point, I'd like to see this method done, if it works consistently without contams, I agree it would be a great method.
