agar, here's how you do it

The medium will be sterilised in an erlenmeyer flask.


dry ingredients are poured into the erlenmyer flask. 8 grams of malt extraxt 8 grams of agar. This is good for 400 ml of medium.



Water is added till 400 ml



It is stirred to mix the ingredients. The flask is capped with a piece of tyvek and this is capped with a piece on aluminum foil.



The flask is sterilised for 30 minutes at 121oC.



The agar is shaken and the foil and tyvek are removed once the flask has cooled down a bit. If the agar is too hot there will be a lot of condenstation on the inside of the lids. If it gets too cold it will solidy.
This should take place in a clean environment like a glovebox or flow cabinet.


Immediately dishes are poured. They are stacked to minimize condensation on the inside of the lids.



Once they have cooled down they are ready to use.

I have a friend who used to inject the actual agar w. spores from a syringe. Meaning, he would stick the needle inside the agar and inject the spores in there. I haven't really heard of this method since then, anybody know what this would do differently, or if it would work well??

dont think the mycellium could breathe "inside" the agar. There is a recent post about just dropping 4 drops, one in each quadrant, on top of the agar. this should work. the other day a friend squirted I believe .25ccs in his plates, maybe more( believe he first said 1cc until I said that was horrible). I think that is why they didn't germinate. Though his room was badly sealed
Also he did it around the edge of the plate 'cause he saw spores streaked that way on some old video. He's not doing that anymore. Personally i believe it'd be best to leave the syringes to the jars. That if you're going agar then it'd make more sense to go w/prints and tissue.

Out of curiosity,

why do you add the tyvek layer? or cotton layer (as I've heard more often) for that matter?

Some dude i know funnels his pre mixed/simmered agar into a flask, then just makes a nice cap that goes 2 inches or so down the neck of the flask wit Al foil. He just pulls the cap off and pours. I never understood the reasoning behind the cotton, or in your case tyvek. Please enlighten me!

As the jar cools down the air is sucked in from outside.
Filter lets the air in while keepeing the contaminants out.
