Mycelium probs...
    #1000329 - 10/28/02 05:00 PM (14 years, 1 month ago)

Ok, I ordered GT syringes. I inoculated 10 jars and waited like 3 weeks. And after that I couldn't notice any mycelium grow at all. So I decided to try again and throw all jars away, except one that had grown a little mycelium. I didn't notice this until I was on my way to throw it away and opened the lid. All the other jars were already thrown away but I saved that one. I boiled 10 new jars and inoculated them too. Now I think it's passed 2 months more and no growth in the new jars. But the "old" jar is less than 10% colonized, 2 months!!?? It's not supposed to take this long, why is that? And why wont the new jars colonize?
*Worried*

I have had somewhat the same problem when I didn?t shake the syringe up.
I would say shake up that syringe real good, make sure you inoculate in 4 points around the jars, and shake between each new innoculation.
Also remeber to keep them at about 85 degrees or so.
Good Luck

Since you had almost no growth I'd guess that your water content was off and that your cakes were either too dry or wet for mycelium growth. Also temperature plays an important role. You want to keep you're jars right about 83 degrees. I know the FAQ and such say 85 but the jars will produce some of their own heat. Also in the beginning stages of growth a certain c02 concentration is beneficial, but after you see some aggressive growth you want to take the tape off and flip the jars. Too much c02 will stall the growth completly. Good luck

Review your procedures. The substrate may have been overcooked, had too much or too little water, or been too hot when you innoculated (killing all or the majority of spores). You should also be incubating at a temperature in the low 80s in total darkness ideally. Did the jars you discarded have a sour smell? Its possible that your substrate contaminated with bacteria (not always visible). If you are sure of your sterile procedures and all the above are also correct, you might want to try a different spore supply.

I used the regular tek (3 parts verm+1 part rice powder+1 part water). Boiled for about half an hour. Cooled overnight, the inoculated like 2-2,5 cc. The temperature is room temperature. Sorry about the bad spelling  I come from Sweden you know

Boil the jars for an hour at the least.
I don?t think half an hour is sufficient.

Like the last dude said, 1/2 hour is not nearly enough boiling time. Your jars may have fallen to contams. Also the PF tek is 2 parts verm, 1 part BRF, 1 part h20.
