Mycelium on a petri dish, center of growth off colored?
    #9629957 - 01/18/09 03:20 AM (15 years, 2 months ago)

Hmm, I used a MS Syringe to innoc some petri dishes. Now looking at the growth.. the center appears to be an off color. maybe very light brown, or tan.

Is this normal? I'm including some photos.

Thanks

I am assuming it can vary on the type of fungus your growing. I have heard different species have different colors according to the books I have read. I haven't seen different colors, but I haven't grown many species.

why would you use a spore syringe on a petri? It is probably a contam. Take a better picture. cell phone camers are not suitable for this hobby.

That looks like cladisporium or possibly penicillium.  You need to start making transfers within days of inoculating petri dishes in order to isolate healthy mycelium away from the naturally occurring contaminants.

Thanks alot guys.

RR-- Thanks I did not know that. Once I transfer the mycelium to another dish, do I need to continue transferring periodically?

or should the myc be able to colonize the dish quickly enough, since the spores don't have to germinate?

Toss that dish out, it's ruined.  You NEVER want to fully colonize a petri dish.  Transfer before contaminant spores form, so you don't ruin your mycelium by having contaminants all over it.

Tahoe - I'm inoculating a petri dish with a syringe to get used to using them. And the fact I don't have a live culture to start with.

I just want to see the difference between selecting faster growing mycelium to inoculate my grains with.

I'm a noob, don't make fun.

:P

Oh I see, Yeah I tossed it out already. I'm about to pour some more agar for round 2.

Thanks for the infos.

using a spore suringe on a petri is a horible idea. It is not a good way to get used to using petris. If you must use the syringe you need to make sure of a few things first.

Clean syringe- most syringes are not clean enough to use on petris or lc's

Pour your agar when it is almost completely cool- you do not need any excess moisture swirling around your petris. This liquid will pick up good and bad spores and bring them across the whole petri.

Use less than a drop of the spore syringe. Push the plundger on the syringe so that you see  a drop of water just starting to form on the tip of the needle. Touch the needle to the petri. Do not let it drop unless it is a very very small drop. You do not want the water to sread out at all. As stated above, the water will drag shit around your whole dish

So are you saying to plush the plunger just enough to where a droplet forms at the tip of the needle and then streak the tip with the small amount of water across the dish?
