Re: Hello, can I sterilize media filled glass petris in the microwave?

I wouldn't reccomend it. Heating agar excessively will cause the sugars to caramalise and this will inhibit growth. Pressure cooking is the best way to go.

Ooh, yeah, pressure cooking. We all know how cool it is in one of those, right?

Glass petris? You lucky...

Yeah. Nukes can do anything pressure cookers can. Do a few bursts of 3 or 4 minutes on full power, then give it a minute to cool. Leave the whole lot in the nuke overnight to cool down. That way, any air that touches the media should be air from the inside of the nuke, and therefore sterile.

Let us know how it does, ne?

I will try it tonight and will let you guys know. Thanks for the input

I am a lucky SOB since I got 50 glass petris

Burple, I just noticed your post, sorry I didnt reply before. I did it. What heppened to Moe is prolly becuase he didnt have pyrex or some other heat resistant petri's.
Now, I did a few experiments with this - the major problem I found was over drying of the agar. M/Ws work by sending waves which move the water molecules faster and faster thus causing them to warm up, until vaporization.
This causes the agar to get quite dry.
#1 experiment - I placed a few petris in the M/W and nuked for 8 minutes. Agar became to dry.
#2 experiment - I placed a few petris in the M/W with aa glass of distilled water in the side and nuked for 8 minutes. had a 70% sterlizing succes (maybe this wasnt due to the M/W but to my actions later).
#3 experiment I placed 6 petris inside a rice cooker (its a little plastic pot which goes in the m/w and really heats up, ccoking a large amouint of rice in 20 minutes). I also added a few inches of distilled water in the bottom of the cooker. Nuked for 10 minutes. 100% succes.
#4 experiment -placed a few dishes in the m/w, with a glass of distilled water in the side. Nuked for 4.5 minutes and waited 5 minutes. Nuked for another 4.5 minutes. 100% succes.

Hope this helps.

Later,
tweedy

For your Guidance:

Twenty four plates were put in the oven rack empty and "cooked" for 2 hours at 140 centrigrade.

I made up my MEA after boiling the water for 20 mins.

Let the oven cool to 30 Centigrade with the MEA juice cooling with it.

Poured the dishes as soon as everything had cooled so that the agar didn;t cook on contact. Made mistake with one dish initially pouting while too hot.

The petris are incubating at 80 Farheneit.
2 days and no contam germination.

I didn't use the microwave yet I;ll see how this works because one can do many dishes
with the oven serilization tek (if it works)

keeping fingers crossed!

Well bad news, :frown:

Out of 24 only 5 made it without contams.
I wait on them a while before I use them to make sure.

I'm glad I have an abundance of everything so it works for me this way.

Cleanup is a bitch but what to do.

Personally I am reverting to the old fashion Press. Cooker. Better odds/more hassle but better than cleanup!

el hornomicrondas != good...

Check it out, here is the deal.

Without pressure you cannot raise the temp above boiling. Reason is goes down to all those funky equations from science classes:
PV=nRT hehe... I can't believe I remember that.

The base is, it will NEVER get hotter than 100 degrees celcius, it doesn't matter if you set your oven at 140. Because the process of boiling cools the substrate down to 100 till all the water is gone. When the water is gone, it will heat above 100 celcius, but without water the whole damn things pretty useless.

Issue #2 with el microndas:
Duration... microwaves cannot provide the same duration as pcooker. As someone duely noted it dries overly much. The key is having the right temp for the right amount of time.

Oh yea, and make sure to eat some shrooms. Oh, and don't disobey the golden rule of shroom growing:
Don't grow shrooms while on shrooms. At least it never works for me no matter how many times I try it.

Hehe...
later,
ootw.
