Liquid Culture Question

Can liquid culture still form in a Karo solution if its environment is air tight? Like if the jars with the karo and silicone seal sucked the spore solution/mycelium water right out of the syringe?

It can but not very good. The vacuum will suck the solution into the jar so only have the amount you want to go into the LC in your syringe. When you start to draw it out if it grows you will have to inject the same amount of air into the jar to the amount of LC you want to take out.
That being said I would break the seal and start over.

no way . its fine with the vaccume in the jar. and when you got to suck it out, the vaccume isnt so strong you cant suck up mycelium. a filter would make it grow thicker. but many have done LC closed in jars with only injection ports.

The times I have done that the suction has so great that I couldn't get it out without it trying to suck it back in. and when injecting the spores it just sucked every bit of solution right in fast. The sores germinated but didn't grow as good as the jars I do without a vacuum.

I just read a post where blutjager grew lc with the same method thick enough to "choak on" in qt jars. alot of people use smaller jars for storeing lc,s of their favorite strains. only injection ports. things should be fine

you expect the vaccume and hold the plunger as to regulate how much is going in. dont let it suck too much. pull it out! hahaha same for getting the mycelium out. expect the vaccume and pull as much as you want. if you let go it will suck it back into the jar. but just pullthe needle out while keeping the liquid in the syringe.

if you lleave enough air space then there will be plenty of air to get a decent amount of lc. obviously gas exchange will help things though.
i believe that if you dont tighten the lids and tighten them after pressure cooking and letting them cool co mplelty this will eliminate the vacumm. but

lol if you pc then tighten lids as they cool it creates the vaccume as well.

Yep leave it a little loose and tighten AFTER it cools.

its been 2 days, I dont have them heated, but I'm not really seeing any growth. is this normal? Maybe I made the liquid culture too thin? the water is yellow, but not syrupy thick. I'm new to this LC stuff. I dont see any contaminants growing though, but nothing is growing. *nervous*

this is what your looking for in early signs of a healthy lc. you want to be able to see through the liquid you dont want the liquid cloudy at all.
lc's will turn out just fine in a vacum but ime it's alot easier to work with them if you had some sort of means for gas exchange ie a whatman or a syringe filter so that its easier to inject spores without injecting a whole syringe worth and getting mycelium from a finished lc which you basically almost need 3 hands to do so the vacum doesnt pull it all back in.

looking a little lighter than that, but basically the same. I tried shooting some with straight up spores (what i had left over) and then I took some mushroom tissue and diluted peroxide and sanatized water/glass and made some mycelium water? (wrong term?) and injected a few with that. Some have vaccum seals going on, others have a poly fill filter. For the one i used a botched spore print. I cleaned the jar with alcohol, and I thought it had all evaporated, so i threw a cap in.. and then I came back a while later and the jar was alcoholy reaking and the mushroo had dropped the spores into a few drops of alcohol. The mushroom really looked bad. But i figured shit, squirt some cooked up karo water in there and seal er up, cant hurt. I left the cap in there (probably a bad idea, but I just wanted to see what would happen)
then I looked at the syringe and realized that there was still a good portion of spores stuck in the bottom of it. So i changed needles and sucked up karo water into that, and then took off the needle in a glove box, wiped it down in alcohol (the syringe end) and wiped down the syringe cap (that little screw in plastic nub thats there before ya unscrew it and toss the needle on) and screwed that back on there.
I'm just doing a lot of experimenting here I guess. Thanks for all your help guys. This is the only "web community" I really "belong" to.

I started an LC this past Saturday and saw just a little growth after about 3 days in one of my jars and say growth in the other jar on the 4th day. Just be patient and it will happen.

I created a filter syringe to equalize the pressure. What I did to make the syringe was take a small amount of a coffee filter and wrapped it around some poly-fill and pushed it into the syringe body (not too tight tho) and let the air in.

soo if the liquid culture IS NOT cloudy, and theres white stringish masses that aren't some odd coloration (whitish yellowish from the karo or honey colored water) that means im on the right track?

basically looks like thick polyfill is growing in the bottom of the jars. The liquid is clear. Its what I'd think mycelium would look like growing in a liquid.
I dont see any discoloration, it all seems uniform in color, nothing looks green, the liquid is yellowish. Sorry about the lighting, that was the best way I could make it show.
