Mycelium stopped growing - picture included
    #8743685 - 08/08/08 11:56 AM (15 years, 7 months ago)

Okay so about a month ago, or longer, I inoculated the jars after using the pf tek method, following all the steps exactly. There was a problem, before I started innoculating I stabbed the syringe very deep into my thumb and bled everywhere. I cleaned up best I could and my husband finished. He sterilized the needle and used a glove box and for the first two weeks here is what grew:

in case the first link doesn't work:
http://img411.imageshack.us/my.php?image=img0032hh9.jpg

and it hasnt grown any more for the last 2-3 weeks. Ive tried jars upside down, loosen lid, theyre in a 75 degree house...

It doesnt appear to have contamination, it just looks wet. Tell me what you think.

Oh and we ended up with enough substrate for only four jars, and as you can see they are not wide mouth, but still half pint. Couldnt find wide mouth. Another thing- my husband used the whole syringe on all four jars, like 10-12ccs, so could be wet.

What should I do with my situation? I dont know how to case, should I learn and do it now? I planned on them growing straight from the cakes.

Well it looks too wet, and your mix might be uneven.  Dunno, I think you might have to restart.

Oh and those are widemouth.  The others taper in.  Most quart jars Are NOT widemouth, most half pints are widemouth.

restart.. looks like it wasnt mixed right, the water content is off, and it look like you got something else naughty in there.. throw them out and start over.. its not worth your time to wait for them to recover..

Yeah you kinda 'uckfeyed' it... Just to let you know, when mixing the brf it goes as follows.

1. 2 cups verm
2. 2 cups water
3. Mix untill consistanst and evenly wet.
4. Add 1 cup BRF and add little at a time and stir, then repeat until all the BRF is added little every few stirs.

-Looks like you added the verm and BRF... THEN added the water. If not that then you added too much water.

ugh...I can sympathize(correct spelling?). I stepped on a needle yesterday. Went RIGHT through my foot.
Start over with the jars.

I also just noticed, do you have any microtape over those holes?  I think you have contams as well.  (The brownish stuff).

Just barely took it off for the pictures. I thought it needed more air. Should I ditch the jars right now or can I get any use out of this experiment at all? It just seems like such a waste

really? I have always done it like a chef would.. mixing dry ingredients first to evenly distribute them, then add wet ingredients..

Toss them out.  They're contaminated with bacteria.  The tell-tale sign is the 'milky' appearance where the substrate is pushed against the glass.

Any way you mix the flour and verm is fine, as long as you get it well mixed.  With most preparations, I mix all the dry ingredients first, and then add moisture.  However, with brf cakes, the flour is so much finer than the verm, it tends to sink to the bottom of your bowl and not mix in well if you do it that way.  Roadkill, who did the pf tek demonstration on 'Let's Grow Mushrooms' uses the verm and water mix first, followed by slowly adding the rice flour.  His method definitely seems to work better for pf type cakes.

However, that isn't what caused your problem.  The bacteria was caused by exposure to bacteria after sterilization.

My bro had a cake with some of that grayish looking stuff on it, but the myc overtook it it was so small of an area. He's fruiting that thing in it's own little bitty container. It's probably still got some bacteria inside though.

So I tossed em. I actually did watch the 'Let's Grow Mushrooms' videos as my guide so if you-Roger Rabbit star in those videos then it's like chatting with a celebrity ha! Well how do I prevent fkkin it up next time? Should I just buy a pressure cooker or was stabbing my thumb the problem.

A pressure cooker would be key, it'd help you keep your jars much more sterile.  Did you not pressurecook your substrate?  By the last post you made it sounds like you didn't PC it.

If you steam sterilized for at least 60 minutes then you can rule out the sterilization of the substrate.

RR's video recommends 90 minutes which has worked flawlessly for myself.

BRF jars do NOT need to be PCed. That is one reason a lot of beginners start with cakes.

Your jars are definitely contaminated. Look at the leading edges of the myc. Notice how it is not fuzzy. That is where the myc ran into a "WALL" of contamination per say.

I would say to work on your sterile techniques during inoculation.

Also 10 cc's per pint jar is way excessive and is going to blow your moisture content.

Those are wide mouth PINT jars.

This definitely would have helped the contaminants along.

Better luck next time.

Also look into doing a LC so that you are not burning up so much $$$ using MS syringes to knock up your jars.

A properly done LC will provide you with LOTS of syringes, for LOTS of jars.

Yeah I was going to ask if she's gone with the same time steaming as PCing.
