So I just received my shipment of Salicylic Acid... I was sitting here running thru the numbers of what needs to be added, and there isnt much info on this particular agar additive...

So I was looking at the ratio's added of other products to a Quart of agar mix...
Gentomyicin Sulfate is added to agar by 1/20th or 0.05 Grams to 1 quart of your favorite agar mix, to create an antibacterial agar.

Some have used Hydrogen Peroxide in ratios of up to 4% or 40ml to 1 Quart of Pre-sterilized agar mix... H202 will break down in the PC... to create an Antibacterial Agar.

Snake Venom has been used in Agar to attempt to create "Hybrid" and/or Cross Species, some interesting experiments have been done, and interesting hybrids/cross-species/interspecies-hybrids created using this method... But there is no info on how much should be added to the agar mix.

Salicylic Acid has been used in the medical field in concentrations up to 35% and over the counter in concentrations up to 17% for the use of Wart removal and a short list of other things.

So after looking at all of this, plus basic sugar ratios and whatnot used in agar, I believe that this additive would work well at a concentration level of up to 1/4 or 0.25 grams Per Quart, and still be useful to my needs, tho i may begin at a lesser concentration and make some observations under the microscope during some simple experiments to try and find the most useful concentration, without harming the mycelium's growth, yet still allowing for the deconstruction of the cell walls...

Maybe someone with more experience than I can help me out here, because my figures could be way off...

Salicylic Acid is also used for the treatment of Excema, Psoriasis, and Acne, In concentrations ranging from 1/16th of a teaspoon (???weight???) to 20% of the total contents... so i dont think that 20% or 8 grams per quart of agar would be right...

Now im gonna have to break out the scale and weigh 1/16th of a teaspoon of salicylic acid... Any ideas how much 1/16th of a teaspoon looks like??? cause that sure is one whacked ass method of measurement!

Weigh a Level Teaspoon and divide by 16 i guess...

I have no experience with it, so unless someone else does, you'll have to guess at the amount to use.

If you're trying to cross cube strains, it might help with dikaryotic/dikaryotic crosses, but won't be necessary if you're wishing to cross monokaryons from different strains.  They should readily cross if they're compatible.

Yeah i was planning to use it in regards to Dikaryotic crosses of cube strains.

I will bust out some experimentation and log my results... not a big deal, just a small side project;)

If I remember correctly, 1 tsp = 5 mL.  If you're talking about water, that's also 1 gram.  So, basic assumption is 5 grams/tsp.  So, 5grams / 16  (or 1/16th of a tsp) is 0.3125 grams (or 312.5 mg).  You do a search on google and I'm sure you'll eventually be able to find a density for salicylic acid to do a weight correction to make a better estimate/assumption of weight.  But that should at least give you so place to start!  Good luck and keep us posted!

well thankyou for that... but i already weighed it and you are so very close to correct that those are actually the numbers that i am basing my estimates on...

I am going to try 2 ratios... 1 @ 312.5mg and another @ 625mg

Im sure either would work to do what i want, however at higher concentrations it may be dangerous to the Mycelium it self... so like i said...

To the experiments!

it's nice to keep all pertinent information on a topic cross-linked or in one thread.  It's not like we have to talk about your topic if you include here. Discussion of his method will continue if there is still something to talk about.  If not, that has nothing to do with you including more information.  (Plus, you may end up saving him a ton of work if it's a working, proven method.)

The best thing about that niacin pdf is that it gives you a recipe for the niacin agar. Takes a lot of the guesswork out of the process.

I would suggest starting with 2-5%.  That concentration should cause some effects without being overly toxic.  I've never used salicylic acid in agar before, but your best bet would be to do some experiments and find the highest concentration that still allow acceptable growth.

Be sure to post your results as this is an interesting topic.

According to some online calculators that equals out to be somewhere in the 2-5 grams per quart for a 2-5% concentration... does that seem right to you?

Ok.
2 experiments about to go underway...
1a. Salicylic acid concentration of 2.5%... Acid will be added first, to look for degradation during Sterilization Process.
1b. Salicylic acid concentration of 5%... Acid will be added first, to look for degradation during Sterilization Process.
2a. Salicylic acid concentration of 2.5%... Acid will be added after Sterilization in front of Flowhood
2a. Salicylic acid concentration of 5%... Acid will be added after Sterilization in front of Flowhood

Ph will be tested in agar solution of 2.5% and 5% before and after Sterilization, while also testing base Ph of agar solution without the Salicylic Acid.

I will be pouring them into small 60mmX15mm petri dishes, allow to cool for 24 hours in front of flowhood before inoculation occurs.

If I seem to be missing anything, or you have a particular method you would like to see used/tested let me know.
