Anyone notice a difference between natural water(stream, spring, whatever) and tap water when preparing a substrate for cakes?

Water that comes out of a tap is not artificial. It has chlorine added, and if you live in some hellhole, fluorine to.

You can distill your own water quite easily, I will release a pictorial tek sometime in the future but for right now all I can tell you is...
Buy a cheap kettle, remove the lid that makes it whistle.
Measure the opening of the mouth (fairly certain this is universal for kettles)
Take your measurements to a local hardware store and find a hose that can withstand abit of heat and will fit tightly around the kettle spout, take a coffee filter or two and pull them down tightly over one end of the hose and either use a elastic band or tape to hold it in place, next, feed the end with the filters to a pre-sterilized jar with a opening just large enough to fit the filtered end of the hose in forming a tight seal to the jar (this can also be achieved by drilling out a hole in a mason jar lid)
Set your kettle to high and wait, you've now distilled your own water and additives are no longer a issue...

you can just pour the tap water into a vat and leave it overnight for the chlorine to evaporate out. I doubt the extra minerals in there are going to do be any concern. Especially if your growing it in horseshit.

Most of my PF jars have been done with tap water without any problems that weren't the result of my own mistakes.
I've also used water from a creek/stream in North Carolina with no problem, but I've never had a problem with tap water either, so you should be fine.

Actually, if you look into bottled water content, you will find it is nearly identical to tap, with all the added chems that are in tap water, maybe smaller amounts depending on the brand.
Distilled water is the only "pure" water there is.

It all depends on the local water supply. In some areas it's not suitable. Also, water from wells often has too many minerals to work well. If in doubt, use spring or distilled water, or if you have a well, run it through a filter. In other areas, the local water supply is fine. I make everything with tap water: cakes, grains, substrates, misting, etc. I only use distilled water for agar. RR

Hell hole? Flourine? Are you calling almost all of the major cities in America a hellhole? That the correlations between the dramatic reductions in tooth decay in the past 30 years since the introduction of fluoridation in the water supply a result of some hellish plan?
Oh, by the way, I use tap water. Philadelphia tap is slightly hard, so it helps to stabilize PH in whatever I do. I boil the tap water if I need something closer to sterile.

Agreed. I grew up with flouride in the water supply, and I'm now a grandfather but have only had three cavities filled in my entire life.
At any rate, most US tap water is suitable for mycology. RR

There's numerous reasons why I feel sorry for anyone living in north america. Industrial bauxite effluent being shipped off as sodium fluoride and dumped into the water system is one of them. It works alright on tooth enamel, but there's no benefit to drinking fluoridated water, and it's no substitute for toothpaste. And who here doesn't brush their teeth?
http://fluoridealert.org/
I regard fluoridation of water in much the same light as mercury being used as a preservative in injections. There's absolutely no need for these sort of toxic substances, and I'm fortunate (as is the majority of the world) not to have to worry about fluorine in my drinking water.
I still use a filter for the chlorine though.

Actaully the only reason we have Flouride in our drinking water is because the aluminium production copanies in this contry had some scientists find some small benifit to tooth enamel strength so they could get rid of this by-product by putting it in our drinking water supply system. Look it up, it is crazy shit. They forced this stuff in large quantities on vietnam soldiers in the 1960's only to find out they were having overwhelming numbers of soldier with enamel difficiancy. Now they are also linking the water supply version of flouride to early puberty in young children. Ever notice how kids seem to be growing up quicker and quicker? The closest city to where I live just launched a huge protest last year to get flouride removed from thier water supply. Everyone had black skull and cross bone signs in thier front yard all year. This is how I came to look all of this information up. If you don't believe me just google "flouride early puberty" Don't believe everything the tube of toothpaste tells you. These companys get paid by the aluminium plants to get rid of this shit, and the only reason they have gotten away with it this long is because it "hasn't been clearly proven to cause any negetive side effects. So because they arent sure if it's bad for you, it must be ok for.

Well, except for Hooper Bay in 1992. Damn, America sometimes reminds me of a sunnier version of Scotland, with more religious folk, and even more shafting.
