Contamination setbacks..
    #683526 - 06/17/02 08:48 AM (21 years, 9 months ago)

Well, I finally got what I thought was a clean culture going from some half-assed prints I took a year ago. I've been using honey water, and adding h202 every now and then to keep it what I assumed to be clean and isolated from contams. But the jars I innoculated ended up with contams growing right along with the mycelium, and it's really knocked my confidence down.
Every time I think I'm getting somewhere, I end up back at square one. I guess this's one of those reminders of how much of a threat contams really are.
Anyway, Is there any advice on what I can do to get some clean healthy mycelium going again? I was thinking of adding a diluted bleach solution to the culture, but I'm wondering if it's even worth my time. Is it even possible to revive it? Thanks in advance.

I don't think you can save it but you can avoid contams in hte future by, of course, working more methodicly and in a clean enviroment. I doubt this is what you are looking for though.

*Sighs*
It's sad, because I have a small jar full of very rhizomorphic mycelium right now. I can't comprehend having to throw it out, but maybe it's for the best. Definately a lesson in not cutting corners..

Personally I wouldn't mess with honey water.. but that's may just be me.
Right now I am doing grain to grain transfers.. it's easy.. and it works like a charm.

Yeah, I'm looking forward to getting into that. I don't mind the PF tek, but it's getting a bit old. However I don't have a pressure cooker at the moment, so I'm doing what I can without one. (which isn't much )

nO way, the honey tech or any similar malt extract works much better inmo that multigrain.
Firstly I get growth after just 1 day after innoculation.
And it strong growth.
Think about it,
a) with multigrain you are exposing the grain to the air, in a glovebox no problems.
But then the same applies for honey water.
b) the grain your using might have been contaminated but being covered in contam and beeing beeten by the mycelium you`ll never know it.
So you do your grain transfer and wham, contamination.

I`ve never had problems with honey water, using a clone or just spores.
Maybe you just need to spray your work area or use a glovebox

Well, I added a squirt of 1/9 bleach solution to the honey water last night, and plan to make at least one more attempt. This time around I'm definately going to be more sterile. I'm thinking maybe I didn't sterilize the syringe/needle enough, because the mycelium I've got growing in the jar looks extremely healthy & vigorous.
Really wish I had a pc right now.. I could be working with agar and stuff.
Anyway, any more suggestions are welcome.

works well hippie but not necessarily needed.
I only use a bit of bleach to help prolongue the life of the jar.
So far I`ve kept a jar for 4 weeks and it`s still transferrable.
and that`s without h202

Of course, np.
Would definately be great when you can't use agar to isolate a dirty print. Probably hopeful thinking, but hey, no one ever got anywhere sticking to one proven method..
