Rye Jars Growing Way Too Slow
    #6734202 - 04/01/07 08:26 AM (17 years, 1 day ago)

I inoculated these quart jars back on 03/07/07 with a newly made LC. It is now 04/01/07 and 3 of the jars are showing minimal growth and 2 of the jars are showing no growth whatsoever. One jar was lost to green mold (I know what happened here). The jars are being kept in a TiT at around 80 degrees. They were each inoculated with 4cc's of very healthy LC. Anyone have any ideas?

Either your jars are too wet, or your lc is bad. I don't think they are too dry considering you

. If you inoculated with a viable lc you should have colonized jars within 6-10 days, depending. My jars colonize in 6 days(qt's)with my Lc's.

Positive it's mycelium. I believe the jars may just be too wet as there is quite a bit standing in the bottom.

If you have standing/pooling water in the bottom of a wbs/grain jar.
I suggest you START OVER.
As the jars will fubar/contam/stall sooner or later.

Water in the bottom is a sure recipe for failure from bacterial contams.

So you either added too much water, or it leaked in during sterilization. (the answer would depend on how you prepared the jars)

Whats in those jars by the way? WBS? Rye?

Water content of a wbs, rye, or pf jar is sooo important, you gotta get that part right.

If you don't see any growth in a jar in the first 2 weeks, (1 week for LC, you should see it colonizing in the first day or so) something is wrong, and from my exp its almost always bacterial.

How do I get the water content right? I'm having issues with jars colonizing slowly and not so well. I suspect moisture issues.

after cooking the grain make sure to rinse (with hot water)and strain well.
better yet, dump the grain into a container (i use a food mixing bowl for small amounts) and blow dry with a hair drier.
when all the grains no longer feels wet or sticks to your hand, or a loaded spoon when "upended" load your jars.
make sure the jars are dry before loading.

What I usually do is soak the grain for 12-24 hours. Then I dump it into a strainer and give it a thorough washing. After which I'll let the grain sit in the strainer for a couple of minutes or immediately start loading the grain into jars.

I never cook my grains beforehand. Also, I'm not so sure about drying the grains so much before PC'ing. My jars seem quite high and dry as it is.

Well that depends on what you're doing wrong!

Grains don't have to be blow dried, some ppl do it, but they just need to be drained right. After soaking you have to drain the grain till no water drips off the bottom of the colander. The other possibility is water leaking into the jar during PCing. If thats the problem, just use less water in the PC! You only need a few inches in the bottom, thats it.

Ahhhh...there's the problem! gotta let them puppies drain for 30-60 minutes...Shake the strainer up and down...When no more water drips off, then you load and PC.

Righteous!

Does this also apply to WBS?

You can also use this -- WBS/GRAIN, a simple test, BEFORE you load jars & PC by Agar.

Titmouse and tarnation, now I know why I'm not getting very good results.... Shit, and all this time I thought it was because of LACK of moisture.

Oh well, my new syringe came yesterday, this time I'll get it right!

I just feel stupid now - So if there's ANY extra humidity in there, it's a bad thing? I've got beads of water here and there. I was always afraid of PCing for too long, and the kernels look like they're kind of cooked, you know?
