Monokaryon Identification

I was curious if it's possible to identify a dikaryon versus a monokaryon culture on an agar plate WITHOUT a microscope. For example, if I started a plate with one single spore and another plate with exactly two spores, would I be able to differentiate between the two without a microscope? (assume the two spores are compatible and form a dikaryon culture)

I got the impression that a dikaryon culture would be much more vigorous than a monokaryon culture. I thought they may be visually distinguishable (if started at the same time, same medium, etc)?

Perhaps with a magnifying glass you could see this: Monokaryotic mycelium tends to look more like mold. That is, the mycelium is curly and wiggly, while dikaryons tend to be straighter. Monokaryotic mycelium also has a lot more side branching of the hypahae, but you won't see that without a scope.

Yeah, I have to agree. There are visual differences but you can't be absolutely certain without looking for clamp connections. But if you had two suspected compatible monokaryons and you placed them in the same petri dish and allowed them to grow together, they should show obviously visibly thicker growth after combining.
