re incoculation
    #6500143 - 01/26/07 06:25 PM (17 years, 2 months ago)

i recently injected my jars with golden teacher and b+ from spores101. in the past 2 weeks my b+ jars are taking off with lots of growth. the golden teacher have not done a thing. only the moisture from the syringe is staying. my question is i have another syringe that contains some pinkbuffalo. what would my chances of being sucessful if i just injected the pinkbuffalo over the unsecessful golden teacher, or should i empty all my jars and redo the substrates and start all over?

worked with golden before, took a long time to gow as well, give it another week, meanwhile i'd mix up another jar in addition to a myc. liquid culture, and inject both at the same time. You're jar will take off and you'll have some liquid culture later on. But i wouldn't reinoculate in the same jar. if your teacher isn't growing there must be a reason.

Be careful not to throw off the water content by shooting too much spore solution since you've already inoculated once. I'd wait and see if anything happens. If there's something with that jar causing it to not grow then you're just gonna be wasting spores. Make an lc with what you've got left and then play with the rest.

Make some new jars. Don't risk wasting your spores.

i say give the teachers a little more of a chance..ive noticed them to be a very slow colonizer both times i have grown them..give it a little longer and if nothing happens just dump the jars and start over..no need to chance waisting good spores

I agree with a combination of the two suggestions present here. I think you should wait a little while longer. My novice experience with Goldent teacher has been a long a patient one. Takes a while to colonize complettly. Secondly, i wouldn't reinoculate. I would pitch the old ones that weren't working and start some new ones. Well actually i would start some new ones now and just keep the others growing too unti lthere is sign of myc. or contamination. You might push up the temp a few more degrees too. but what you have should be adequate for growth.
