Prefered agar mix?
    #576951 - 03/12/02 01:53 PM (22 years, 1 month ago)

PDA,MEA, MYP, MPG?
a la Stamets, Oss & Oeric, or your own special blend?

I switch back and forth between PDA and MEA. I get the PDA from a local lab supply company and I get MEA+antibiotic from fungi perfect. The PDA I get is a bit more expensive than the MEA combo, but it is also a lot better quality.

PDA is very easy to make.
i find it works the very best. MEA didnt work much for me whatsoever.

boil 4 or 5 big potatoes in a liter or little more of water (you'll see why in a minute)

chop the potatoes up with the skins on
rince them a couple times, then boil em for 1/2 hour

strain the potatoes away and SAVE the water they were boiling in

top this water up to 1L (4 cups)
while its stil in the pot on the stove add in;

10g of dextrose powder
15g agar agar
1.5g nutritional yeast
throw in 5g of malt extract if you feel horny

let hte agar agar liquify into this mixture

then pour it out into a jar that you can pressure cook (make sure its down about 2" from the top so it doesnt bubble over the top of the jar into your PC.
(this shit likes to foam and bubble)

pc 'er for at least 30 mins. i go 45 to be honest.
make sure the lid isnt on tight on your containers.

if you have small pressure cookable 1/2 pinte wide mouth jars, you can put the amount you want right into them, and cook it like that.

just make sure that if you're gonna store the agar, that you pressure cook it first..
it stores well in quart jars

MEA. Bacteriological agar and LDME. (light dry malt extract). Sooooo easy.
Some "potato flakes"- ghetto mashed potatoes in a box- can be added in small amts to "mix it up", if you need to alter the mix for "dexterity".
I have even added BRF and powdered millet for kicks.....
Just do not stick with one formula for tooooooo many plates.
Agar is so easy and REWARDING. Bottom line- when you isolate a GREAT "CLONE". Nothing will make you happier......well....

It has to be noted though, that most of these agars are not suitable for propagation of higher funghi.

Thank you all for the feedback.
cookiewhore~MYP is malt/yeast/peptone and supposed to be very good "for long term maintenance of desireable cultures" according to Oss & Oeric's book. To one liter you add 7 grams of malt extract, 1gram peptone or sytone ("commerical brands of a protein hydrolysate"), half a gram yeast extract and 15 g agar. Stamets also has a recipe for it with a slight variation. MPG is malt/ peptone/grain the recipie is in The Mushroom Cultivator. I believe this would probably be one of the best 'cause if I rembember correctly, in addition to the peptone/sytone rye and a splash of yeast is used. I'll post stamets recipies for you tomorrow or Mon. as it is in my journal in a locker in Antartica
Anno~ will the MPG be good for p. trops. or, better yet, what would you reccomend for them?

could someone please point me in the right direction?
like what sort of place i'm likely to find it in.
thanks for any response.

here you go...
MPG:
20g tan malt
5g ground rye
5g peptone or neopeptone(or sytone~ REMEMBER, THEY'RE JUST BRAND NAMES FOR THE SAME PROTIEN)
2g yeast (optional~but I'll bet Stamets doesn't leave it out)
20 grams agar

morlahere,
just click on the sporeworks add in this forum ($20/100grams)or another vendor if you don't want that much you could pm dimitri. He's probably still selling in catered amounts.

Oh, heres Stamets MEA:
20g tan malt
2g yeast
20g agar
Stamets says, "Avoid dark brewer's malts which have become caramelized. The malt that should be used is a light tan brewer's malt which is powdery, not sticky in form." But from what Oss & Oeric says this doesn't mean you can't use a syrup. Just relates to when using powder(alot neater than syrup).

My foavorite is PDYA with ever changing flour additions(wheat, millet, rye, soy, amaranth,corn...) or additions of dung extract for Panaeolus species.

I have to side with feito.  Bacteriological grade agar and a mix of ghetto tatos or MEA

The bacteriological is simply a higher grade of agar.  It solidifies "better" as in it gels harder and it cooks up nice too!
