Liquid Culture Questions

1. I have read someone saying they put their liquid culture in a TIT to get it up to 80 degrees, and I have heard someone else say leave it at room temperature. Which should I do?
2. I read that you should keep the LC going at around 100-300RPM if you are using a stirrer, but I have seen pictures where the water is swirling into a tornado but mine only does that at really high RPM. To what RPM should the stirrer be set?
3. I used a 4 percent dextrose/malt solution and filtered it with two coffee filters between two 20min 15psi PC sessions. Then when I turned the stirrer on and turned it up till I got the tornado going a white froth like on a beer formed at the top. Is this usual and or a problem?

You want the stirrer low, I turn it up like a vortex only when the LC is getting thick and only for a few minutes.
Room temp can work for some I had success with it.
I would PC @10psi for 20
No need to turn it that high when you start it.

1.) I always kept mine incubated. I had a big incubator so it wasn't really an issue. Anything near 80's is good.
2.) I swirled once a day maybe, and I'd swirl it fast enough to generate a pretty steady tornado. It never hurt my mycelium.
3.) The foam was probably just caused by the disturbance of the water. I used honey for my LC's and the top wasn't exactly frothy but it did have a cluster of bubbles.

I did not have luck with Malt/Dext until I weighed it out,
5g each per 2cups
3days from 1cc of LC, room temp and indirect light, 24/7 stirring
I have another going from 1cc multispore, day 2 there are a few strands going.
A better filter set up for around 30-40 and that I just love using and there is no sediment what so ever,

Yeah, I made the mixture by weight. First I filled my jar with the amount of water I wanted in it then I transfered it over to a lighter plastic cup to weigh it. I took this weight and divided by 96 to figure out what one percent would be. I then multiplied this number by 4 to determine the amount of dextrose/malt extract I would need. Then divided by two to figure out how much of each I needed. This should then be 96 percent water and 4 percent dextrose/malt extract right? I used a scale that is acurate to 0.1g
Where did you get that strainer from?

This = less than 3% mixture, 4% would be 9+g of each for 16oz water. The only thing I?ve noticed is growth stops when the sugars are consumed, so I assume that 4% = more growth.

Shazoom that is just under the 4% mark at around 3.8-3.9%. I remembered agar saying that he had the best results with a just under 4% solution. I was wondering if the solution you listed is about the mark I should be shooting for then? Also would straining the solution take out enough to bring it down to under the 4% mark since you are getting rid of some of the sediment?

After running the stirrer overnight I noticed it got way to hot. The temperature was hanging around 100-105 F. I went out and acquired another stirrer that I'm certain won't overheat. My question is should I keep the LC that I just started even though it hit those temperatures for a brief period? It was on the plate for maybe 12 hours. Is this long enough to damage the spores, or are temperatures that high only bad for mycelium that has started to grow?

You'll need a Glass Flask, Pump, Buchner filter and filter paper and an adaptors, they sell them as a 'kit' for 30-40.

Anyone have any ideas as to whether or not I will be able to continue using this LC or if I should just give up and start a fresh one when my new stirrer comes in?

keep it going and see, might as well. if not, make another. after 12 hours at 100+, i'd think the jar is shot.

Well I'm not sure how quickly the temperature rose but it was probably pretty fast. However I guess my biggest question is can spores with stand that kind of temperature. Because I doubt it had time for the spores to have started mating and creating mycelium.

anything over 100 and you can pretty much give it up.
I keep mine right at 80-82 and all is good.
I spin 100-300 and only turn it up a bit when it gets clumpy to let the stir bar shred up the myc.
vortex is bad!
I pc at 15psi for 20 minutes, that is plenty.
hmmm...what else, , , good luck I guess.
lc is the way to go, you'll be very happy you decided to switch over.
