is this petri contamined? (pic)
    #4310099 - 06/18/05 02:39 AM (18 years, 8 months ago)

something alien and strange is growing in my petri dishes.
like a contam, it starts from the sides and moves towards the centre, while the myc starts from the centre and goes to the sides.
unlike contams it's not green or red or purple or any fancy colors: it looks transparent, just a little darker than the agar medium, which is brownish. maybe a bacterial contamuination? do they look like this? it is my first petri contam.
soon this thing and the myc will meet.
what shall i do?

my first petri too

it looks like contam, but maybe not if it looks transparent. I had some dishes that got patches of that too. maybe fermentation of the agar? looks like you successfully transferred once to this plate already. You may as well cut this one up, innoc and transfer to another petri. The myc growth will likely be effected when it comes to the brown region.

i tossed it, doing 3rd generation today even if petris of the 2nd generation aren't completel? colonized. most of them have the same kind of suspect contam. will make 8 new petris out of the 2 best old ones.
just asking myself how many generations should i do before transferring agar to grain?

as long as there is clean myc in the middle, you should be able to salvage an infected dish. I cut up one dish with green into 12 jars and only got one jar with green.

do you use peroxide in your agar? that's the next step for me, but I'm lazy so I've put it off until this weekend, thus the overgrown petri that I showed

i added a little h2o2 to the agar mix this time for the first time (3rd generation). 1.5ml h2o2 to 200ml agar mixture (0.75%), added just before solidification. i will transfer a "healty" part of myc from the previous generation in each dish. my choice for a healthy mycelium follows these rules:

1) myc from a non contamined looking dish over myc from a contamined looking dish
2) rhizomorphic myc over cottony myc
3) faster running myc over slower running myc (running from the centre to the edges of the petri dish of course).

in this way i'm hoping to isolate a fast, rhizomorphic and contam resistant substrain. only after that i will test its fruiting capabilities.

i guess we're going off topic though

I only have 1 second generation. It was cottony from my second petri from the gills of shroom to a third petri. The third is all cottony but some of the a separate batch look to have some rhizo growing. I started last weekend so I'm working on figuring out what to do this weekend.
