autoclave help please
    #371104 - 08/13/01 12:11 PM (22 years, 7 months ago)

hey y'all. i have access to autoclaves and i know how to use them. I was wondering if anyone out there knows the proper temp and time for sterilizing 1/2 pt jars of brf/verm and for 1qt jars of rye/millet. I don't want to overcook the grains or torch the flour, these things can get very very hot and have high pressure. I was thinking that I should use 121C and liquid cycle (as opposed to the dry cycle for solid objects), but for how long?
thanks a bundle,
heathadelic

Have you a pressure guage? At 15 psi, 20 - 30 minutes for brf/verm. For the quart jars 1 hour.

At 15 psi the temp is 121?C.

I would go 45 min with the pf jars and 1 hour with birdseed or rye.
Liquid cycle.(steam)

I have used my autoclave for pf tek/brf and recently for finch/brf.
I have done test runs with indicator strips inside the jars and also.. cut open substrate after
to make sure the brf didn't crystalize. You can go ahead and run it up all the way
to 260degrees F and high pressures of 121pounds/ 25-30 MIN. REALLY! BUT YA GOTTA
ADD EXTRA WATER FOR EACH CAKE or the cakes will be dry. About 1/8 -1/4cup of water
for each 1/2pint jar. Drippy is good...actually I made up a batch of substrate according
to recipe..and then added alittle over an 8th of a cup of water to the jar.
I learned the first few times..the hard way.. too dry cakes. I also tested the jars with temp
indicator strips with and without tape over the holes
.My last cakes were finch seed
done with micron majick discs with the added water. They colonized in record time...
Some were still slightly dry and I dunk tekked all but one. The one I didn't came out of the
jar moist, and that was the one I added the most water to.. .all are pinning now.

Also.. I think the finchseed (hawk recipe) came out better in the autoclave than
brf/verm pf recipe not only because of the added water but more airspace.
The brf pf recipe..the substrate is more compact. I had 5 jars in per load, standing upright.
If you have autoclave tape.. use an indelible marker and write on the tape the amount
of water you used. Tape it to the outside of the jar to keep a record. I also mark the
tape later with the strain for id too. The tape won't come off. k..

I was wondering if those filter discs were autoclavable....
Do you think I would experience the same amount of water loss if I just used the jar lids with the holes covered with autoclave tape instead of the filter discs?

The filter discs are autoclavable. I really don't think there will be much difference
in the water loss. Do them both ways and see which one you like best.
I just remembered.. I put the filter disk on and then the jar lid over that.
As it turned out when I went to inoculate I found that trying to get the needle through
the holes and then through the filter was a wipe out. I took the lids off. "They" said
that the discs would seal to the jars.. Well.. they didn't. I held the disk in place and stuck my
needle through it. I didn't like that at all.. so I ended up taking the damn disc off and
inoculating (carefully) down the sides of the jar, replacing the disk, and putting another
disk over it for a double layer of protection. I also wear gloves, mask, and use a hepa filter..
soooooo .. it's all a matter of experimenting and practice.
