Will spores germinate in agar?

Germinate and grow myc, that is.
Could I throw some spores onto a fully prepared agar dish and expect them to colonize it after a while, rather than taking a cutting of my baby?
Just curious.

spores germinate not in agar but on agar, they form mycelium, thats called a tissue culture.
you usually dont throw them on but wipe them on, using an inoculation loop for eample of a sterile needle.
anyway you shall work very sterile since on agar anything will germinate, not just your spores. peace ohm

I have found quicker germination when working with agar if I break the surface of the agar with my loop and place some spore in the break when I'm streaking, this is usually if not always the place germination first occurs also. I've also noticed the myc was noticably more vigorous coming from this area. Just a tip I learned long ago and works very well for me. GL

So basically you plant some spores beneath the surface of the agar. If agar germinates spores/bacteria/etc., I'm assuming that you could introduce mycelium to the agar as well. Nifty... How long does one usually keep an agar culture for?

Not to long..they dry out. You can definatly keep them for a few months..I've kept them up to about 2 I think..somewhere around there. I dont know if I stored them well..just wrapped in clingfilm in a cool dark place..
One ended up fruiting...I cant seem to find pictures though..dont remember if I took any..

Sorry to dredge up an old post, but I was doing some searching on the subject of spores to agar and came across it.
I was thinking an alternative to scratching the agar would be to streak the spores out and then just add 3 or so ml of top-agar (a less concentrated agar) or for that matter, just another layer of regular agar. This would provide the best rehydration potential since the spores would be 'submerged' in agar.
Of course care must be taken that the agar isn't too hot when you pour it, but this is a common technique in bacterial culturing so I'm sure the more resilient mushroom spores would survive the brief exposure to heat.

that sounds like a pretty good technique as long as temperature is carefully monitored, however, spores suck. I find it much easier/successful to use mycelium when innoculating agar.

Live myc transfer to agar begins colonizing much faster than spores.
Spores take time to germinate, then go.

Ya, I don't doubt it...but if you've only got a spore print to work with, well, then you've only got a spore print to work with.
Anyone know about how long it takes for the spores to germinate and show growth?

if you only have a spore print to germinate. Do a MSI inoculation to grain, grow 1 flush and clone a few of the biggest mushrooms, then you already have isolated strains and you may select the phenotype.

But if spores are unclean you could and up with nothing. Had that experience. Started agar 5 days ago still no germination, will wait and see. I think agar is the key to micology, playing with MS is more about luck. Had pore result of MS and got tired of failing.

Taking a cutting though is a clone and is better genetics because it will only be a handful of strains, which all most.likelu are fruit bearing vs m.s. where half the strains do.not.produc emushrooms
