my first agar project

hi guys, i read lots of recipes for preparing agar plates....
i want you to tell me if the procedure is correct,i won't tlak about quantities of each ingredient. just the procedure.
ingredients: Agar agar, Potatoes, Dextrose,petri dishes

procedure:
i melt the potatoes in a pot and boil them in water i then only keep the liquid.
I put this liquid in a bowl and add agar agar and dextrose and mix them.
I sterilize this mix for 20'
I let it cool slowly....
and i pour into the plates...

please tell me if i am wrong somewhere in the procedure
questions:
1)dextrose i can find in a drug store in powder form.is this what we want?
2)if i have more liquid than the petri dishes,how could i keep it alive for a few weeks/months?

I've heard that agar can last months regardless...
But i don't know to much about agar,

Never done the homeaide agars , but sounds about right .
Personally would pc for 40 mins , but that is me .
Yes if you make too much , just store in the fridge,in a glass botle with narow neck, with a very good airtight lid , and when ready to use again just put in pot of hot water till easy to pour , can store in fridge easily for a few weeks to a month .

PS TMC has a couple of recipes for making own agars .Also remeber when making own agars , one should periodically check ph of the mixture .

trip, how in fact do you do agar if you don't make your own? Have you boughten over 200 pre-sterilized petri dishes with agar in them?

And if I wanted to spend that kind o money , yes one could buy pre poured petris with agar in them , but I add water , pc , and pour .
And to you ,for now the name is MR Tripndicular , not trip !

ps , just for you , I may ask friend to take pics (no face included) while I make , pour , and inoc , tissue transfer etc ..... WITH NO GB OR FH ! Will take some time to prep and make sure friend is clean enough (lots of greasy hippy friends  :smirk:) to help me ! Why oh why do I feel the need to proove myself ?

sounds pretty good. dice the spuds up small and keep the skins on - then cook em 20 mins and pour it through a sieve, keep the liquid.

add water to the volume your recipe says, and add the dextrose (yes, the stuff in the chemists) and the agar (chinese shops sell lots of this). some recipes also add yeast, or yeast extract. pc in a glass container with a chunk of polyfil in the top, and don't fill it all the way up, as it can boil and bubble. 20 minutes should do it.

open the lid when it stops hissing but is still hot. to germinate spores, it's now ready to pour; but to grow mycelium from mycelium (ie. wedges) you want to add some peroxide after it cools below 140 and before it cools to setting temperature (around 125). you need a 'candy thermometer' for this. don't worry - it takes bloody ages to cool down when you're waiting for it.

the good thing about peroxide agar is that it does keep for ever. i make up litre batches of PDA, pour all the plates i want, then stuff polyfil in the bottle neck and just leave it on the sideboard. stays for months, until i want some more plates - this time i just put the solidified agar-bottle in the pc and give it a little longer, maybe 40 minutes, so it melts. of course, it needs new peroxide each time, as it denatures above about 145.


agar is fun - look forward to trading wedges with ya, hamurabi  :laugh: :thumbup:

if you wanna do some nice agar,
id advise you to try my beer agar project aswell.

click the link in the signature,
it goes all about peroxiding and how to prepare it in detail.

and you dont need malt extract.

This is best way to make and store agar IMHO

Insertion of syringe needles to act as a vent ...


Needles in place ready to stick in the PC ...


Of course one pulls the needles prior to storeage , those are just to act as vents while in the PC . And yes that is just water in there , this is an example of a way to make and know stays clean in the fridge , and no worries with water soaking cotton or poly etc .

I find it funny that there is a beer-tek for agar...

I use beer to promote moss growth... Just raw beer! Works great! Actualy, the starch and yeast method has been used for hundreds of years. The ancient chinese would pour all of thier water used to clean rice, all over thier gardens and stones/statues. Within weeks-months, you would see massive moss growth, thick and bright green. Never needs mowing, looks better than grass, cleans the water that passes through it, and stays green all winter!

They would do the same with any fermented beverages/foods (Grain alcohol, not really beer!)
