H2O2(Hydrogen Peroxide) Mycelium Syringe
    #3430868 - 12/01/04 12:29 AM (19 years, 3 months ago)

Here is a H2O2 mycelium syringe using ideas from the petri peroxide agar tek. After searching I found nothing mentioning the amount of peroxide to use in the myc water or how to go about it.
Heres first steps with pics.

1. Supplies:
-Get 500ml of distilled and boiled water(cooled down in container with sealed lid)(tap water has chlorine which kills myc and destroys H2O2)
-Get a bottle of H2O2 (3% from store)
-Get a syringe (with a screw on/off removable needle)
-Get a shot glass
-Boil some water
-A flame
-Plastic Gloves
-A flat-head blade(xacto kits have blades that are flat on the end, not pointed)
-Peroxide Agar Petri with good mycelium growth

2-Sterilize your syringe Needle, Flame it! Sterilize your syringe, suck up boiling water into it, and let sit for 5 minutes, squeeze all water out into sink. Then suck H2O2(3%) into syringe (suck as much as you can) and let sit for 5 minutes. Squeeze out into sink.

3-Sterilize your shot glass, pour boiling water into it, over fill it, and let it sit for 5 minutes. Dump into sink. Then after, fill it with straight H2O2(3%) and let sit for 5 minutes) Dump into sink.

4-Suck 3ML of H2O2(3%) into syringe and inject all into 500ml of water. Stir around with needle a bit.

5-Suck up a FULL syringe of the (500ml)Water+(3ml)H2O2 into the needle. Naturally you can't get it all, 500ml was used to keep the numbers even and not fractional.

6-Open Peroxide Agar Petri and take sterile flathead blade and lightly scrape off the top of the agar to remove mycelium ONLY. It will look as if your removing almost nothing at all. DON'T DIG into the agar at all, just scrape the mycelium off the top of it, it will happen very eaisly.

7-Hold flat blade over shot glass, and take needle(with the H2O2+water) and wash the flat blade with myc on it into shot glass. Repeat this for more mycelium in your shot glass water. When done, squeeze all water+h2o2 out of syringe into shot glass.

8-FILL syringe lid with H2O2(3%) and put the lid back on the empty syringe. (filling it isn't easy, tap lid while filling it) UNSCREW syringe needle and set needle aside.

9-Take needle-less syringe and put into shot glass full of mycelium+H2O2+water and quickly suck ALL of shot glass contents into syringe.

10-Screw syringe needle back on.

(if you find your mycelium is too thick to inject with needle on, when sucking up into needle-less syringe, plunge it back out quickly into shot glass to break up mycelium do this a few times if needed, you can then put needle on and plunge it out again into shot glass, and suck it up with needle on to break it up even more)

Use this syringe immediately, as its prolly NOT a good way to store mycelium. H2O2 breaks down under light. I don't know, I have never tried this to store long term Myc syringes. I also don't know so far if this method is going to be too "dirty" and introduce contams. If so, then I might suggest adding even more H2O2 to the 500ml of water.

sounds interesting. how does the mycelium react to all the h2o2? i would imagine like you said, it wouldnt be a good way to store it.

small amounts of h2o2 can be beneficial to myc as it converts it to o2.

well, considering its using the same concentration that the peroxide agar tek uses, im guessing the mycelium only gets hurt a little bit, as it gets used to it. But the petri tek only exposes the myc to the surface, and not even all sides, like the myc H2O2-water does. I guess the only way to find out is try it, innoc somethin, and watch.
I don't think the myc will grow much at all, as theres no nutrient in the water-H2O2. Mabee this is a good way to store??

The concentration that the tek says is very small, to every 1000ml of water add 6ml of H2O2(3%)
Above, I halved the amount of water and H2O2 so that one didn't have so much useless Water-H2O2.

This works, the mycelium used, recovered, and is now growing white fuzzy growth around the "sick" looking mycelium that was scraped from the petri and sucked up into the H2O2-Water syringe. The NEW mycelium growing on the medium looks very normal. Will report back after even more growth shows.

Looks great, the H2O2 mycelial syringe is a total success. No contaminants at all, with a fully colonized substrate using this method.

NO GLOVE BOX OR OVEN TEK WAS USED DOING THIS. JUST SMOOTH QUICK WORK.

This worked really well for clearing up cobweb in grain jars @ 10-20 % colonization. Minor setback 2-3 day stall then recovered . Thanks for the info fella's

rule of thumb if anything in your jar isnt mycelium throw it away and start over.

good luck i guess

No . i prefer not to submit pics of my Elm Oysters lol.

I will however submit some photos ( courtesy of someone else ) a experiment of this sort with PF-Tek affected by the green stuff. All I can say for now is ...pretty impressive ! . I'll post when he get's final results.
