How much FAE does agar need? parafilm vs micropore vs ketchup cup
    #26941597 - 09/18/20 12:50 AM (3 years, 6 months ago)

hey there. I did try and search this but didn't get a satisfactory answer. As it stands, Im using petri dishes sealed with parafilm for the air exchange - it seems to be a safe way to go. But I see a lot of people using ketchup cups with no air exchange. What gives?

Does anyone here have experience with ketchup cups? do they need a hole with micropore tape?

And does anyone use micropore on petri dishes? I guess my overriding question is in the title. How much air exchange does the mycelium need when it's growing on agar if any?

Parafilm seems to be the gold standard. what do you think?

It can take over the entire surface with only the air in the dish.

The sauce cups probably dont meed the filtered hole because they dont seal well enough to crush under the pressure

Stick with the parafilm.

Why would anyone want to wrap their plates in micropore tape? Peeling that shit off in order to make a xsfer sounds horrible. What suggestion is next? Duct tape?

And whatever you do... tho some may say its awesome (its not)... keep any cling/saran wrap in your kitchen.

Saran wrap is for leftovers not culutres.

And thats the first i've heard of "ketchup cups" being used to seal plates. I dont like the sound of that. Sounds like another gem from the reddits.

Thanks very much. ok that's good to know. was thinking ketchup cups might be a cheaper option for doing lots of transfers.

Cheers

Some people have been using those pre sterilized (not exactly but pretty clean from the manufacturing process I guess) ketchup cups but it looks turrible to me. I tried some pp5 containers once and I hated it. Petri dishes with Parafilm all the way.
